Expression And Meaning Of 5-lipoxygenase And Leukotriene In Aorta Of Atherosclerotic Rats

Objective Through the model of rats atherosclerosis, discuss the meaning and pathogenesis of 5-LO/LTs role in atherosclerosis.Method①Seventy-five normal male SD rats were divided into five groups randomly,they were A:normal control group(n=15);B:atherosclerotic group(n=15);C:montelukast sodium 1mg group(n=15);D:montelukast sodium 3mg group(n=15) and E:simvastatin group (n=15).②normal control group was fed with normal diet,the other groups were fed with high cholesterol diet for 14 weeks in order to found the model of atherosclerotic rats;③montelukast sodium 1mg group,montelukast sodium 3mg group and simvastatingroup were treated with montelukast sodium and simvastatin for 4 weeks,then the rats were killed, blood lipids and 5-lipoxygenase , leukotriene B4, C4 in blood and aorta were measured in five groups,and aorta were observed by Hemotoxylin-Easin staining and transmission electron microscope.Results Blood lipid in atherosclerotic group,montelukast sodium 1mg group,montelukast sodium 3mg group was higher than normal control group(P<0.01),it was not different within these three groups(P>0.05);Blood lipid in simvastatin group was lower than atherosclerotic control group,montelukast sodium 1mg group,montelukast sodium 3mg group(P<0.01),and was not different than normal control group(P>0.05)。Blood and aorta 5-lipoxygenase,LTB4,LTC4 in atherosclerotic group,montelukast sodium 1mg group,simvastatin group was higher than normal control group(P<0.05),and was not different within these three groups;Blood and aorta LTC4 in montelukast sodium 3mg group was lower than atherosclerotic group(P<0.05). Atherosclerotic group intimal proliferate obviously, and montelukast sodium 3mg group,simvastatin group intimal was thinner than atheroscler- otic group.Conclusions The expression of 5-lipoxygenase , leukotriene B4, leukotriene C4 in blood and artor of atherosclerotic rats was abnormal;montelukast sodium can reduce LTC4,restrain atherosclerosis,but can not reduce blood lipid; The role of 5-LO/LTs may be a new target to prevent atherosclerosis。...
