BACKGROUD:Cervical cancer with highly morbidity and mortality is one of the most common gynecologic malignancy, which has a high incidence in Xinjiang. It has increased year by year and tends to be younger nowadays. Surgery and radiotherapy are the main conventional cancer treatment which can achieve a better efficacy in the early cervical cancer. But for the patients of advanced cervical cancer often died of tumor recurrence and metastasis these treatments are not satisfactory. For the patients of cervical cancer who lose the chance of surgery or radiation,palliative treatment is preferred. We usually give priority to chemotherapy in clinical. For the chemother drugs (cisplatin,carboplatin,5-Fu and gemcitabine) commonly used in cerical cancer have large toxicity and be easy to have resistance, it seriously affect the patient's therapeutic effect and quality of life. So,looking for the drugs and combination chemotherapy regimens with high efficiency is important. Studies have shown that Pleurotus ferulae(PF) may be a promising anticancer drugs. However, the mechanisms of resistance of polysaccharide for cervical cancer are not clear entirely.This is the research of the anti-cancer polysaccharides ferulae mechanism was discussed.Objective:To observe the growth-inhibition and the apoptosis induced by polysaccharides from Pleurotus ferulae (PFP),cispitain(DDP) and their combination on murine cervical carcinoma U14; To investigate the mechanisms of PFP in the treatment of cancer.Methods:The mice with U14 were randomly divided into four groups:Model group,DDP group,PFP group,PFP+DDP group. Intervention were given different drugs for 10 days, the changes in tumor volume of mice, tumor weight and tumor inhibitory rate. were measured. Apoptosis-related protein levels of Bcl-2 and Bax were observed by immunity histochemistry technology. The U14 cell apoptosis index (AI) was assessed by TUNEL.Results:Beginning of the experiment, no difference in tumor volume in each group, the Seventh day after the experiment began, the intervention group were lower than the tumor volume growth rate of model group, the tenth day, the intervention group tumor volume was significantly smaller than the model group (P<0.01), PFP+DDP group tumor volume was less than the DDP group (P<0.05);The weight and of tumor in model group were higher than that in other three groups. The weight of tumor in PFP+DDP group (0.40±0.61)g, respectively, were lower than that in DDP group (0.61±0.26)g.The tumor cell apoptosis in experimental group were higher than those in model group,and the apoptosis index was highest in PFP+DDP group. PFP treament and DDP treament alone or in combination down-regulated expression of Bcl-2 proteins and upregulated expression of Bax proteins.The effect of combined therapy in down-regulating Bcl-2 and upregulated Bax was stronger than that induced by gossypol or radiotherapy alone.Conclusion:1.Polysaccharide from Pleurotus ferulae significantly inhibited tumor cell growth of U14 cervical cancer in mice.2. Apoptosis was induced by Polysaccharide from Pleurotus ferulae on the U14 cells, the induction of apoptosis may be associated with decreased expression of Bcl-2, increased Bax protein expression, thereby reducing the ratio of Bcl-2/Bax in the function.3.Polysaccharide from Pleurotus ferulae combinated chemotherapy drug cisplatin, can increase the inhibitory effect of cisplatin on U14 cervical cancer cells, which relate to the downregulation of Bcl-2 protein and upregulation of Bax protein,indicating a synergistic effect between two.
