Objective: To study the effects of tumor suppressor gene IL-24 on cervical cancer cell line Siha using the shuttle vector pAd track CMV.Methods: The shuttle vector pAd track CMV that carried the objective gene of IL-24 was transfected into cervical cancer cell line Siha. Methyl thiazolyl tetrazolium (MTT) rapid photocolorimetric assay was used to examine the cell proliferation of Siha. Flow cytometry was used to examine the cell cycle and rate of apoptosis of Siha. At the same time, pAd track CMV were transfected into Siha as empty-vector control, wild-grow Siha as negative control.Result:1,The OD values showed significant difference among three groups of cells by MTT assay (F=17.565, P<0.001). Compare to the other two groups, the OD values of IL-24 group was obviously reduced(P<0.001), but no significant difference was shown between empty-vector group and negative control group(P=0.100). The growth rate of IL-24 transfected cells was inhibited,while the cell growth curve of empty-vector group was similar to that of negative control group.2,The apoptosis rates of negative control group, empty-vector group and IL-24 group were 2.70%, 3.18% and 8.83%. The apoptosis rates showed significant difference among three groups of cells by flow cytometry (P<0.05). Compare to the other two groups, the apoptosis rates of IL-24 group was obviously reduced(P<0.05), but no significant difference was shown between empty-vector group and negative control group(P>0.05).3,By flow cytometry, the cell percentage in G0/G1, S and G2/M period showed significant difference among three groups of cells(G0/G1:F=657.980,P<0.05;S:F=213.390,P<0.05;G2/M:F=24.669,P<0.05). Compare to the other two groups, the number of IL-24 transfected cells in G0/G1 period was increased , cells in S and G2/M period were reduced (P<0.05), but no significant difference was shown between empty-vector group and negative control group(P>0.05).Conclusion:1,IL-24 gene can inhibit the growth of cervical cancer cell line Siha.2,IL-24 gene induce the apoptosis of cervical cancer cell line Siha.
