Objective: To establish a simple and reliable cerebral vasospasm model after subarachnoid hemorrhage through auto logus arterial blood injection into the cisterna magna twice in rabbit, after Magnesium Sulfate Iniection was injected into the muscle, we observed the transform of Neuroethologic score and assessed the blood flow rate of basal artery of rabbit and the morphologic change of basilar artery with hemotoxylin and eosin staining and Interleukin-17 of blood plasma and expressions of Transforming growth factor-β1 in front lobe following vasospasm. after subarachnoid hemorrhage, studying the effect of magnesium sulfate on Interleukin-17 of blood plasma and expressions of transforming growth factor-β1 in front lobe, in order to explore deeplier pathophysiological mechanism that magnesium sulfate can relieve Vasospasm and improve Neural function,providing more theoretical basis for the treatment with magnesium sulfate was used in clinical.Methods: After establishing auto logus arterial blood injection into the cisterna magna twice.,we choose 40who is Vasospasm by examining with TCD in 60 healthy male New Zealand white rabbits, which were radomly divided into two groups, including magnesium sulfate -group and saline-group,two groups were equally assigned into 5 subgroups according to time pointsld,3d,5d,7d,14d (n=4). On ld,3d,5d,7d,14d, 25% magnesium sulfate (0.3ml/Kg.d) was injected into the muscle in the medicine group every day,the same saline was injected in the saline-group,we observed Neuroethologic score and assessed the blood flow rate of basal artery of rabbit and the morphologic change of basilar artery with hemotoxylin and eosin staining,we examined the transform of Interleukin-17 of blood plasma and expressions of Transforming growth factor-β1 in front lobe following vasospasm..Results: Rabits of two groups neural action begin to be decline from 1d after subarachnoid Neuroethologic score of two groups are not difference (P>0.05), Damage of neural action of magnesium sulfate -group is less than saline-group from 3 to 14,neuroethologic score of rabbit in saline-group begin to be slower compared with magnesium sulfate—group (P<0.05),The average blood flow rate of basal artery is rised from 1d,it is highest on 7d,,then is deline to 14d when vasospasm is existing,in magnesium sulfate-group on 3d begin to be slower compared with saline-group(P<0.05), it was decresed obviously on 7d (P<0.01), basilar artery can be seen that endothelial cells are incompleted,elastic membrance is distorted into lumen, endothelial cells are swelling,two groups are not significant diffrences, the content of Interleukin-17 of blood plasma is rised firstly,i it is highest on 7d, then is deline, magnesium sulfate-group begin to be slower compared with saline-group(P<0.05) on 3d, it was decresed obviously on 5d,7d,14d (P<0.01), transforming growth factor-β1 was rised firstly from Id to 3d,then was deline, two groups are not significant diffrences on 1d,3d,14d, it was increased obviously on 5d,7d (P<0.01)。Conclusion: 1. Vasospasm is appear after subarachnoid hemorrhage through auto logus arterial blood injection into the cisterna magna twice in rabbit 2.The treatment with magnesium sulfate can relieve Vasospasm of basal artery,and improve Neural function 3.There is no significant effect of magnesium sulfate on the pathological change of BA 4.The effection of magnesium sulfate on Interleukin-17 and Transforming growth factor-β1 is one of pathophysiological mechanism that can relieve Vasospasm and improve Neural function.
