Objective :To discover the "YISHENTANG" mechanism of prevent and treatment of DN, diabetic rat models were induced by a single intraperitoneal injection of streptozocin(STZ) to observe the effect of YISHENTANG on cell cycle regulatory protein p27 in the kidney of diabetic rats, while Valsartan as a control .Methods :41 adult male SD rats were divided randomly into two groups: normal contronl group (CON)and diabetic model group(DM). DM group can be made into diabetes rat models by injecting Streptozotocin(STZ) 60mg/kg into their abdominal cavities at a time, CON group can be injected buffer solution with the same volume。The criteria of diabetic rats is: plasma glucose concentration had been higher than 16.7mmol/L for three days after 72 hours injected STZ. Then the diabetic rats were divided randomly into three groups: diabetic control group(DMN),diabetic group treated orally with Valsartan(ARB) , diabetic group treated orally with YISHENTANG(YS). ARB group were treated with valsartan 20mg/kg everyady, while YS group were treated with YISHENTANG 6.5g/kg, CON and DMN were treated with equivalent drinking water. Every rat was weighted a time per week to adjust the drug dosage in the first three week, and weighted a time everyday in the last five weeks. After 8 weeks, we collected 24 hours urine , and weighted rat body, then sacrificed all rats with anesthesia. Blood were cellected from heart to test plasma glucose, creaeinine, urea nitrogen. Detected the expression of p27 in immunity class with the slice in kidney, and have a quantitative analysis with image analysis system.Results Diabetic models was successfully induced by injection of STZ. Body weight: CON group was significantly greater than all DM group(P <0.05), there was no significantly difference in all DM groups.Renal relative weight of rats: the renal relative weight in all diabetic rats was significantly greater than that of CON(P <0.05), there was no significantly difference in all DM groups. Plasma glucose: plasma glucose in all the diabetic rats were significantly higher than CON group,while there was no significantly difference in all DM groups. Plasma creatinine: there was no significantly difference in all groups .BUN: BUN in all the diabetic rats were significantly greater than CON, while there was no significantly difference in all DM groups. 24 hour urine volume: 24 hour urine volume in all diabetic rats was significantly greater than that of CON(P <0.05). 24h urine total proteins: 24h urine total proteins in all diabetic rats was significantly greater than that of CON(P <0.05),and there was no significantly difference in all DM groups. 24h urine albumin :24 hours urine albumin protein in all diabetic rats was significantly greater than that of CON(P <0.05), ARB and YS were significantly less than that of DMN(P <0.05),while there was no significantly difference in YS and ARB. Expression of p27:expression of p27 in DMN group were significantly greater than CON, ARB and YS(P <0.05),and ARB and YS were significantly less than that of DMN(P <0.05). Correlation Analysis: 24 hours urine albumin protein was significantly positive correlative with expression of p27.Conclusion: 1,YISHENTANG can reduced the 24 hours urinary albumin,and decreased the expression of p27 in the kidney. It is probable that the effects of YISHENTANG on treating DN and reducing albuminuria's excretion would due, in parts to decreasing expression of p27 in kidney.2,YISHENTANG have a similar function with ARB in reducing diabetic rats urinary albumin, and decreasing expression of p27 in kidney.
