Wound is caused by environmental factor and internal factor such as obstacles of blood circulation, which was often accompanied by destruction of skin integrity and loss of tissue. Chronic wound is a kind of wound that the healing process lasts more than 2 weeks. Chronic wound is often caused by diabetes, varix, arteritis, arteriosclerosis, ischemic disease and nervous system disease. In China, the incidence rate of diabetes, ulcer of lower limb and decuhitus are increasing every year, in which acute wound turn to chronic wound because of local blood circulation failure. Many clinical investigations have proved that this kind of chronic wound could lead to disability, in which local blood circulation failure is the most important influential factor. Thus, seeking new drug that can accelerate wound healing is of great importance. Chuang Yu Sheng Ji Powder is a new drug based on traditional prescriptions and manufactured by the Fourth Military Medical University, which can facilitate wound healing. Aim to analysis the therapeutical effect and potential mechanism of Chuang Yu Sheng Ji Powder on chronic wound, the present study utilize diabetes animal model on rat and ischemia animal model on rabbit to form two different kind of chronic wound to dectect the therapeutical effect and potential mechanism of Chuang Yu Sheng Ji Powder. Compare the wound healing time of animal model cured by Chuang Yu Sheng Ji Powder with that of blank control. Investigate the expression of VEGF, HIF-1αand Ang1, which closely related to angiopoiesis, in tissue sample of chronic wound by Real time PCR and immunohistochemistry. Detect microvessel density by immunohistochemistry assay using poly clone antibody of CD31 or CD34.Results showed that the wound healing time of animals with diabetes treated with Chuang Yu Sheng Ji Powder, Recombinant Bovine Basic Fibroblast Growth Factor and blank control were (21.3±2.1)d, (23.2±2.4)d and (26.8±2.8)d respectively, among which the difference was statistically significant (P<0.05). While the wound healing time of animals with local circulation failure treated with Chuang Yu Sheng Ji Powder, Recombinant Bovine Basic Fibroblast Growth Factor and blank control were (22.6±2.3)d, (24.7±2.5)d and (27.2±2.8)d respectively, among which the difference was statistically significant (P<0.05). Real time PCR and immunohistochemistry showed that the expression of VEGF, HIF-1αand Ang1 were up-regulated in animals treated with Chuang Yu Sheng Ji Powder, compared with animals treated with Recombinant Bovine Basic Fibroblast Growth Factor and blank control. And the microvessel density of tissues from animals treated with Chuang Yu Sheng Ji Powder was significantly higher than the other two groups (P<0.05).The results above showed that Chuang Yu Sheng Ji Powder could accelerate chronic wound healing through the upregulation of VEGF,HIF-1αand Ang1, which were closely related to angiopoiesis.
