Preeclampsia (PE) is a pregnancy specific disorder, which happens after 20 weeks of gestation. The incidence is about 9.4% in China. Clinical features of PE include high blood pressure, proteinuria, edema, which may cause serious complications. PE is one of the most cause of maternal and fetus mortality. A number of factors have been thought to contribute to PE, including Placenta shallow implantation, hypoxia, extensive damage of vascular endothelial cells, genetic background, Mother-to-child immune imbalances. MicroRNAs (miRNAs) are short ribonucleic acid (RNA) molecules, about 19-25 nucleotides long. MiRNAs are post-transcriptional regulators that bind to complementary sequences on target messenger RNA transcripts (mRNAs), usually resulting in translational repression and gene silencing. miRNAs have been proved to participate in development and varieties of diseases, especially in early development, cell differentiation and apoptosis. Pineles BL proved that some kinds of specific miRNA increasd significantly in PE patients'placenta, which suggests that miRNA may play a role in the pathogenesy of PE. In this study, we compared the expressions of miR-19a in placentas from normal and PE patients. And this may provide some evidences for further study and treatment.Objection:Detect the expression of miR-19a in tissues of placentas from normal and serious PE patients and its roles in the pathogenesis.Methods:1. Detect the expression level of miR-19a in 20 samples of each goups by using real-time PCR.2. Transferred pre-miR-19a into JEG-3 cells to over-express miR-19a and use RT-PCR and western blot to detect the expression of target genes.Results:1. miR-19a increased significantly in placentas from serious PE patients.2. There is no dramatic difference in HLA-G mRNA between JEG-3 transferred cells and non-transferred cells. While protein HLA-G was reduced significantly in miR-19a over-expression group compared with normal group by using western blot.Conclusion:Some specific miRNAs expressions were up or down regulated in PE patients, and miR-19a expression was greatly increased in placenta of PE patients. Results showed that HLA-G maybe the target of miR-19a, and miR-19a may play an important through HLA-G in the pathogenesis of PE.
