Expression Of ING4 In The Tumor Of The Colon And The Rectum

Objective:To investigate the expression of the ING4 mRNA in colorectal cancer,adenoma and normal colorectal epithelium.Methods:1. Collected 40 cases of colorectal cancer specimens (including 4 cases of TNM I,5 cases of TNM II,14 cases of TNM III,17 cases of TNM IV),40 cases of adenoma (including 28 cases of tubular adenoma,12 cases of villous adenoma) and 10 cases of normal colorectal epithelium.2. By reverse transcription polymerase chain reaction (RT-PCR) technology to detect the different levels of ING4mRNA.3. Experimental data processed by T test.Results:1.RT-PCR results showed that ING4 mRNA in colorectal cancer, adenoma and normal epithelium were expressed, the ratio of the densitometry of ING4 and the Densitometry ofβ-actin were 0.37,0.54 and 0.78. There are statistically significant difference in ING4 mRNA expression between colorectal cancer, adenoma and normal epithelium groups (P<0.05).2.RT-PCR results showed that ING4 mRNA in colorectal adenocarcinoma, villous adenoma, tubular adenoma and normal epithelium were expressed, the ratio of the densitometry of ING4 and the Densitometry ofβ-actin were 0.37,0.51,0.56 and 0.78. There are statistically significant difference in ING4 mRNA expression between colorectal adenocarcinoma, tubular adenoma and normal epithelium groups (P<0.05); There are statistically significant difference in ING4 mRNA expression between colorectal adenocarcinoma, villous adenoma and normal epithelium groups (P<0.05); While no statistically significant difference in ING4 mRNA expression between villous adenomas and tubular adenomas groups (P> 0.05).3.RT-PCR results showed that ING4 mRNA in TNM I, II, III, IV colorectal cancer and normal colorectal epithelium,the ratio of the densitometry of ING4 and the Densitometry ofβ-actin were 0.4,0.36,0.22,0.09 and 0.78. There are statistically significant difference in ING4 mRNA expression between TNMⅠ,Ⅱ,Ⅲ, Ⅳcolorectal cancer and normal colorectal epithelium (P<0.05).Conclusion:1.It showed that the expression of ING4 mRNA in colorectal cancer, adenomatous polyps and normal colorectal epithelium was increasing, indicating that ING4 is a tumor suppressor gene, and may be involved in the process of canceration.2. With the increasing of colorectal cancer TNM staging, ING4 mRNA expression is decreased.It suggests that ING4 inhibits the ability of aggressiveness behavior and low expression of ING4 with higher invasion capacity.
