Objective:(1) To study the expression of TK1 in breast cancer.To investigate the correlation of TK1 with ER,PR,p53,C-erbB-2, Ki-67 and conventional pathological parameters in breast cancer.(2) To investigate explor the effection of the expression of TK1 in the metastasis and recurrence of breast cancer.(3) To compare the S-TK1 concentration in breast cancer, in benign breast tissues with in healthy people and to explor its clinical significance.Materials and method:(1) Selected 100 breast cancer cases which were surgical resected in the Shenzhen Second People's Hospital from march 2010 to march 2011.All the 100 cases were biopsied conventional HE staining,and were made clear the organization type, pathological features(grade and lymph node metastasis) and clinical stage of it. ER, PR, p53, C-erbB-2, Ki-67 and TK1 were detected by immunohistochemical method.(2) Selected 101 breast cancer cases which followed up from the Shenzhen Second People's Hospital from January 2003 to January 2006. Selected 15 cases from 34 cases with recurrence or metastasis in 5 years randomly and 20 cases from 67 cases without recurrence and metastasis over 5 years (including 5 years) randomly.All the cases were biopsied conventional HE staining and made clear the pathological grade.TKl was detected by immunohistochemical method.(3) Collected 70 peripheral blood samples of breast cancer,20 peripheral blood samples of benign breast tissues and 10 peripheral blood samples of healthy population. The concentrations of serum TK1 in all the blood samples were measured by enhanched chemiluminesence (ECL) dot blot assay.(4) All the experimental data were analyzed by SPSS 17.0 for windows.Results:(1) The positive staining of TKI was mainly allocated in cytoplasma,partly in cytomembrane. TK1 in 100 patients with breast cancer was 85%(85/100). The positive expression rate of TK1 was significantly different from histological gradeⅠtoⅢand from clinical stageⅠtoⅢ(P< 0.05);and not correlated with that of age,tumor size and lymph node metastasis.(2) The positive expression of TKI was positively correlated with that of Ki-67(r=0.871) and was negative correlated with that of ER and PR (r=-0.374,r=-0.384);and not correlated with that of C-ercB-2 and p53 (P>0.05).(3) The positive expression rates of TKI in group with recurrence or metastasis and without recurrence and metastasis were 93.33%(14/15) and 75%(15/20).The expression of TK1 and the histological grade of tumor were showed statistical difference(P=0.004,0.003) between the two groups. The results of survival analysis showed that expression of TK1 and histological grade of tumor were related with breast cancer recurrence and metastasis.(4) The concentrations of serum TK1 were showed statistical difference (P<0.001) in peripheral blood samples among breast cancer, benign breast tissues and healthy population;and also statistical difference(P<0.001) from histological gradeⅠtoⅢin breast cancer.The results showed that the concentrations of serum TK1 in peripheral blood were positively correlated with the malignant degree of breast cancer.Conclusions:(1) The positive expression rate of TK1 was significantly different from histological gradeⅠtoⅢand from clinical stageⅠtoⅢ.The higher histological grade and TNM stage of tumor are, the higher expression of TK1 is.TK1 is a sensitive Indicator measuring the proliferation of breast cancer cell.(2) The expression of TK1 was correlated with that of Ki-67, ER and PR. The higher expression of TK1 is, the higher expression of Ki-67 is and the lower of ER and PR are in breast cancer.(3) Positive expression of TK1 and high histological grade foreshow a high reccurence or metabasis of breast carcinoma.(4) The concentrations of serum TK1 in peripheral blood samples are rising from healthy population, benign breast tissues, low histological grade breast cancer to high histological grade breast cancer. The serum TK1 is a simple and reliable auxiliary indicator to diagnosis breast diseases.
