Objective:To evaluate the therapeutic effect of BMSCs on the experimental acute liver failure in Rats induced by CCL4.Methods:1. Selecting,separating,cultivating and identifying BMSCsBMSCs were obtained from two rats thighbone under aseptic condition, then cultured and pured by the whole marrow-cultured method. Immunohistochemistry is applied to detect cell-surface marker expressions:CD29,CD44,CD34 and CD45, which help to identify BMSCs.2. Evaluating the therapeutic effect of BMSCs on the experimental acute liver failure in RatsThirty model rats were randomly divided into three groups:normal control group, without any treating; liver failure group, with injection of physiological saline via caudal vein; transplantation group, with BMSCs transplantation of the same dosage, both of which had been injected with 50% CCL4 mixed by paraffin oil, 1.2ml/100g, freshly-prepared, to develop acute liver failure models. Finally, survival rates, liver function and histopathologic examinations were performed, with the results of which to evaluate the therapeutic effect of BMSCs transplantation in the treatment of acute liver failure in rats.Results:1. Immunohistochemical analysis of P3 BMSCs indicated CD29,CD44 positive and CD34,CD45 negative, which presented a brown cell membrane for positive markers.Flow cytometry showed positive rates respectively:CD29(99.3%), CD44(99.6%),CD34(4.3%),CD45(17.1%).2. Rats survival rate showed its increase to 70%, compared to the liver failure group(20%). Besides, the liver function improved gradually, that is, the activity of ALT and AST decreased remarkably. In pathohistology, the degeneration and necrosis of hepatic cells and degree of inflammation were less obvious that that of liver failure group.Conclusions:1.BMSCs,with high-purity and uniform configuration, could be obtained by the whole-marrow adherent culture method.2.BMSCs transplantation via caudal vein is an effective method to treat acute liver failure, increasing its survival rate, improving hepatic function and alleviate the degree of cell necrosis. Definitely, BMSCs transplantation can help to treat experimental liver failure in rats.
