| Objective:To ensure the effective part of Eucommia for anti-photoaging, and conducted the function mechanism and the quality specificationMethods:Eucommia was extracted by ethanol, then Eucommia extract was eluted with gradient of ethanol by macroporous resin(WE,EE). The HaCaT cell and ESF-1 cell model of photoaging were separately builded by UVA and UVB. The effective part of anti-photoaging of eucommia was sured by the index of cell morphology, cytoactive, LDH, SOD activity and MDA content.The mice model of photoaging was did by the effective part. After irradiation, the dorsal skin of animals were analysed by biopsy of skin, SOD activity and MDA content were analysed by kit. The ESF-1 cell model of photoaging was did by EE. The secretions of MMP-1·MMP-3·TIMP-1 Col I·Ela of cultured fibroblasts were detected by enzyme-linked immunosorbent.Result:WE had no significant effect of photoaging. EE could enhance HaCaT cell and ESF-1 cell activeness, reduce the values of LDH, MDA content and increase the value of SOD in culture solution after UVA or UVB irradiating. EE dose can significantly decrease the amounts of MDA and increase the activity of SOD in plasma, the skin of themice has no significantwrinkles, close to the Normal group which 1% EE on the skin. As well it can decrease the amounts of MDA and increase the activity of SOD in plasma. EE have the protective effect on the skin of long-term ultraviolet irradiatedmice. EE have the protective effect on the skin of long-term ultraviolet irradiatedmice. The secretions of MMP-1·MMP-3·Ela were obviously increased and TIMP-1·Col I obviously decreased after ultraviolet A irradiation comparing with the control group. After treated with EE the secretions of Ela decreased and Col I increased.They had significance in statistics.Conclusion:EE can inhibit UV irradiation-induced damage in HaCaT and ESF-1 cells. It's sure that EE was the effective part of Eucommia for anti-photoaging.Skin photoaging phenomenon was effectively improved by topical application of EE,when the effect of topical application of EE was more obvious.Its mechanism is due to its abilities of scavenging oxygen free radical, inhibiting lipid peroxidation, increasing anti-oxidative enzymes, protection of membrane structure, reducing MMP elastin content and increasing collagen content.This study research the mechanism of Eucommia anti-photoaging.And then, provide experical basic for exploitation of anti-photoaging new drugs in Eucommia. |
