Hepatitis C Virus (HCV) is the main pathogen of chronic liver disease, whose infection distribution is worldwide, currently about 170 million infections each year worldwide, and still have 300~400 million new cases in world every year; in our country HCV have about 3.2% infection rates, namely have about 38 million HCV infections,more than 80 percent of whose infections developed into persistent infection,which have chronic hepatitis and liver cirrhosis, liver failure and liver cell cancer relation with chronic hepatitis and liver cirrhosis,liver failure and liver cell cancer, after 20 years infection there are about 10%-20% in patients development to cirrhosis of the liver, after 30 years infection there are about 1%~5% in chronic patients liver development to cancer, At present the mainly arv treatment of chronic HCV infection is peg-intron combining with ribavirin,but in only about 50 percent effective, So whether wo can develop safe and effective preventive therapeutic vaccine became the primary issue of HCV today, but HCV often in the form of quasispecies exists in human body, show different parts with different variation degree,and E2/NS1 has the highest variation degree,which variation may have related with HCV chronic and the disease process of HCV.Like HIV,hepatitis C can be infection through blood transfusions, sexual contact or maternal-infant transmission,so there many co-infectioner,but different route of transmissions have different infection rate, the blood transfusions has hightest infection rate,so there are many co-infections in paid blood donation crowd.ObjectiveResearch the change of E2/NS1 area after merger HIV, found the dinffence between two groups,and provide foundation for merger HIV.MethodsDivide into two groups for all the HCV infections according to whether merge HIV,then genotyped all the HCV,and RT-PCR and SSCPand sequenced for the sequenceing of E2/NS1. ResultsThere were statistical differences between the HCV/HIV merge group and the HCV mono-infection group in genotype of HCV (χ2=35.4, P<0.001),and the HCV mono-infection group has the complex genotype,There were statistical differences between the HCV/HIV co-infection group and the HCV mono-infection group in SSCP strip number (t=2.32, P=0.049).And the HCV mono-infection group has the more SSCP strip.the within-group variation in two group comes from HVR-1,HVR-2,HVR-3; The differences of homology between two group about HCV E2/NS1,HVR-1,HVR-3 has statistically significant; The amino acid sites which have high mutation in HCV mono-infection group to be conservative in HCV/HIV co-infection group.Conclusions1. HCV 1b are.mainly genotypein both groups,but there were statistical differences of HCV genotypes distribution between the two groups.2. HIV can reduce the gene variant of 1b HCV E2/NS1, mainly reflects in HVR-1 and HVR-3 area.
