Endometriosis is a common and popular gynecologic disease in women of the reproductive age and presence and proliferation of endometrial tissue could growth outside the uterine cavity. The main clinical symptoms are dysmenorrhea, chronic pain of pelvic cavity and infertility. Endometrial tissue of Endometriosis is impact by many factors. It has the high invasion and recurrence. Although it's a benign disease, its pattern of growth is similar to malignant disease. EMT harms to women's health seriously and its incidence is rising in recent years.10%-15% of Women of childbearing age and 32%-48% of infertile women suffer EMT.Infertility has many complex pathogens include factors of oviduct,Ovarian,immunity and EMT. Buyalos was the frist scholar expounded the concept of endometriosis- associated infertility in 2000.There are a lot of concepts of the mechanism that EMT how to cause the Infertility. (1) The theory of immunity. The number of the Macrophages is increases in blood and peritoneal fluid of patients of EMT. Macrophages are a negative influence factors for sperm moving, sperm-egg fusion, nidation and fetal development. (2) The theory of oxidative stress. Many endometriosis- associated cell factors could promote the addition of ROS. Oxidative stress may promote the emancipation of cell factors, induce immunity and inflammation. Chronic inflammation changes the physiological structure of cavitas pelvis and many links of gestation. (3) The theory of genito-endocrine secretion. Endometriosis- associated infertility associated with the change of estradiol, progesterone, prolactin and yellow body.Only 10%-15% of women suffer endometriosis who menstrual reflux, so menstrual reflux is the indirect factor of endometriosis. Some scholars believe that endometriosis is a polygenic disease associated with chronic inflammation and oxidative stress. The common manifestations form of the causative agent of endometriosis maybe oxidative stress.With the free radicals, arachidonic acid generate 8-iso-PGF2αby the non-enzymatic pathways.8-iso-PGF2αis a specific product of lipid peroxidation. It without the impact of dietary lipids is the most viable biomarkers of oxidative stress and lipid peroxidation.GSH-Px is an important peroxide decomposition enzyme and important indicator of Antioxidant capacity. Its main physiological function is catalyse GSH participate in oxidizing reaction, clear the peroxide and hydroxyl radicals which be produced by cellular respiration, and lessen the peroxidation of the membrane polyunsaturated fatty acid.Compared with the study of single cell factor or gene, the study of the common manifestations form of the causative agent of endometriosis is more clinical application valuable. The study results of the oxidative stress and its Indicators of endometriosis- associated infertility contribute to the diagnosis and treatment of endometriosis.ObjectiveThe topics collected samples of endometriosis- associated infertility patients and normal control women in Zhengzhou Han people,using enzyme-linked immunosorbent assay (ELISA) technology for 8-iso-PGF2αand chromatometry technology for GSH-Px, for study the expression of 8-iso-PGF2αand GSH-Px in blood serum and peritoneal fluid of endometriosis- associated infertility patients.At the same time,we explore the relations of the levels of EMT and 8-iso-PGF2α,GSH-Px.This examination supplies a new consideration for treatment and preservation of EMT.Material and Methods1 ObjectInpatients with endometriosis and primary infertility or secondary infertility (n=50) accepted laparoscopic operation and the specimens collect at gynecological department of the Third affiliated Hospital of Zheng Zhou University from september in 2009 to february in 2010. The patients met the diagnostic criteria issued by R-AFS,in which there were 10 cases inⅠstage,12 cases inⅡstage,13 cases inⅢstage and 15 cases in IVstage. The specimens were assigned into a group (ⅠandⅡ,22 cases) and B group (ⅢandⅣ,28 cases).Their average ages were (31.701±7.531) years. The results of pathology text of the specimens are endometriosis. The controls were the inpatients without endometriosis and infertility (n=30) at the same time. Their difference in age,weight and menstrual cycle has not statistically significant. All patients had no physical and surgical disease and had not accepted the hormone therapy three months before operations.2 Methods2.1 Sample collectionSerum:Every person be drew off blood 3ml into the test tube before the operation.These specimens should standing one hour in the common temperature, and be centrifuged by 2500r/min 10min. Peritoneal fluid:During the operation, we collect the peritoneal fluid 5-10 ml from Douglas cul-de-sac, which must be centrifuged immediately by 2500r/min 10min.All the types should be conserved under -80℃and waiting for text together.2.2 Empirical methodThe study examine the levels of 8-iso-PGF2αwhich will be examined by enzyme linked immunosorbent assay (ELISA) technology and GSH-Px which will be examined by chromatometry technology in serum and peritoneal fluid.8-iso-PGF2αELISA kit comes from R&D; GSH-Px kit comes from Nanjing Jiancheng. Experimental procedures conducted according to the instruction carefully.3 Statistical analysesSPSS 16.0 was performed to analysis the study data. All the date was express as (x±s).The study data were adequately normally distributed and two population variances were equal at the significant, so two independent samplest test was used. Statistical significance was defined asα=0.05.Results1 Comparison of clinical general informationAnalysis of endometriosis- associated infertility group and normal control group of women clinical data, we can see that comparing the two groups of women in age, differences were not statistically significant (P>0.05).2 The expression of 8-iso-PGF2αin blood serum and peritoneal fluidThe expression of 8-iso-PGF2αin endometriosis- associated infertility group was significantly higher than that in normal control group(P<0.01).The expression of 8-iso-PGF2αin endometriosis B group(ⅢandⅣ)was significantly higher than that in endometriosis A group(ⅠandⅡ) group (P<0.01).3 The expression of GSH-Px in blood serum and peritoneal fluidThe expression of GSH-Px in endometriosis- associated infertility group was significantly lower than that in normal control group (P<0.01).The expression of 8-iso-PGF2αin endometriosis B group(ⅢandⅣ)was significantly lower than that in endometriosis A group(ⅠandⅡ) group (P<0.01).Conclusions1. The oxidative stress exists in the body of endometriosis and infertility women.2. The oxidative stress may correlation with the endometriosis.3. 8-iso-PGF2αcould as a specific biomarker of the level of oxidative stress in endometriosis women and prognostic assessment.
