Spleen Soup Model Of Depression In Mice Colon Tissue Expression Of Sp And Vip

Purpose:The experimental study of depression model mice on behaviour, the first defecating time,substance P (SP) and vascular active bowel peptide (VIP) in colon tissue with affect of Gui-pi Decoction, discusses this decoction's possible mechanism on prevent and control the depression.To provide Gui-pi Decoction for the clinical application an experimental evidence for prevetion and treatment of stomach and intestines dysfunctional disease under the depression.Material and method:40 healthy Kunming mouse, the body weight is 20±2g, are half female and half male. Divided into 4 groups stochastically, each group of 10, divide namely into①Blank group: Conventional raising, does not give any stimulation;②Model group:gives the mold with 21 day of unpredictable mild stress chronic stress (CMS), every day uses the physiological saline to fill the stomach;③Traditional Chinese Medicine group:Makes the mold and use Gui-pi Decoction treat them simultaneously, each 10g body weight with the 0.15ml traditional Chinese medicine fluid (1g/ml including crude herbal medicine) to fill the stomach;④Western medicine group:Makes the mold and gives Fluoxetine HC1 simultaneously solution without capsule, each 10g body weight 0.15ml to fill the stomach.Three weeks later, observe behavior change with the Open-field, test the body weight change, the first defecating time. After the animals are executed, take 1cm long colon tissue above the anus 7-8 cm immediately, make HE staining section, observed under light microscope, use immunohistochemical method to detect groups of mice colon tissue SP and VIP's expression, and analysis the differences between the groups.Results:1. Behavioral score and the weight:①the model group compared with blank group behavioral score and weighing are both lower (P< 0.01), indicating that depression model has been established.②the TCM group compared with model group can improve the mice depression state, the behavior score and weight maintain at a higher level (P<0.01), and compare with western medicine group (P> 0.05) efficacy is close.2. First black defecating time and morphology:(1) after 21 days made molds, model group compared with blank group from first black defecating time significantly shortened (P< 0.01),under the light-microscopy, parts glandular structures not neat, goblet cells reduce and atrophy, the mucosa become thinner, and have inflammatory cells penetration embellish. (2) the TCM group the first black defecating time is close to normal, and compared with the blank group doesn't have statistical significance (P> 0.05), but the model group does (P< 0.01), light-microscopy morphological structure basic normal.3. SP and VIP:model group group compared with blank colon tissue SP express increased (P < 0.05), and TCM group and the model group has significant difference (P< 0.05). Model group compared with blank group colon tissue VIP express reduce (P< 0.05), and TCM group compared with model group has significant difference (P< 0.05).Conclusion:1. Gui-pi Decoction can improve depression model mice depressed.2. Gui-pi Decoction can improve depression model mice colonic motion function.3. Gui-pi Decoction can adjust SP and VIP expression in depression model mice colon tissue.
