Preparation And Identification Of The Monoclonal Antibody Against HoxA1

ObjectiveTo prepare monoclonal antibody against HoxA1 and establish hybridoma cell lines secreting antibodies specific for HoxA1.Methods and ResultsSynthesized immunogen of GST-HoxA1 were used to immune BALB/c mouse。One hybridoma cell strain were obtained, and can stably secret special antibody against HoxA1. Following that, the cells are amplified in vivo of BALB/c mouse. The antibody is purified by repeated precipitation using different concentration of saturated amine sulfate. Moreover, the caprylic acid-(NH₄)₂SO₄ is used to purify the antibody. ELISA and Westen bloting etc are used to respectively identify the McAb. By Westen bloting, the McAb against HoxA1 display strongly specific and high affinity, and has no cross-reaction with carrier protein. The valence of 4D5 may be up to 1:102400.ConclusionThe McAb against HoxA1 is produced successfully.
