Objective:This study aims to investigate the role of Survivin and COX-2 protein in human ameloblastoma and to study the relationship of the two kinds of protein expression between histological subtypes and imaging subtypes in ameloblastoma.Methods:1.64 samples were collected, these specimens of ameloblastoma were divided into seven groups: 4 malignant ameloblastoma; 15 follicular ameloblastoma,13 plexiform ameloblastoma,14 unicystic ameloblastoma,10 acanthomatous ameloblastoma,4 peripheral ameloblastoma and 4 desmoplastic ameloblastoma.11 normal oral mucosa (NOM) tissues and 13 dentigerous cyst(DC) are as normal control. Immunohistochemical streptavidin-biotin perosidase examined expression of Survivin and COX-2 in all samples. The results were analyzed by SPSS 13.0 software wrap.2.60 ameloblastoma pre-operative X-ray film were collected. These specimens were divided into three groups:24 unilocular ameloblastoma,31 multilocular ameloblastoma and 5 honeycomb ameloblastoma. Analyzed the relationship between the results of the two kinds of protein expression and imaging subtypes in ameloblastoma.Results:1. In contrast to negative expression in dentigerous cyst, survivin was expressed in 2 of 11(18.18%) cases of normal oral mucosa tissues,43 of 60(71.67%) cases of ameloblastoma and 4 of 4(100%) cases of malignant ameloblastoma. Comparing with normal oral mucosa tissues and dentigerous cyst, the expression of survivin was higher in ameloblastoma and malignant ameloblastoma (P<0.01). Comparing with ameloblastoma, the expression of survivin was higher in malignant ameloblastoma (P<0.05).2. COX-2 expresses positively in 4 of 11(36.36%) cases of normal oral mucosa tissues,7 of 13 (53.85%) cases of dentigerous cyst,49 of 60(81.67%) cases of ameloblastoma and 4 of 4(100%) cases of malignant ameloblastoma. Comparing with normal oral mucosa tissues, the expression of COX-2 was higher in ameloblastoma and malignant ameloblastoma (P<0.05). Comparing with ameloblastoma,the expression of COX-2 was higher in malignant ameloblastoma(P<0.05).3. There were no relationship between the expression of survivin and COX-2 with age,gender,position or histological subtypes of ameloblastoma (P>0.05)4. The expression of survivin and COX-2 in ameloblastoma correlation positively(r=0.529,p=0.000).5 The expression of Survivin and COX-2 in unilocular ameloblastoma was lower than multilocular ameloblastoma and honeycomb ameloblastoma (P<0.05).Conclusions:1. Survivin and COX-2 may be related with the occurrence process and malignant process of ameloblastoma.2. Survivin and COX-2 may exist mutual adjustment mechanism in ameloblastoma.3. The proliferation of multilocular ameloblastoma and honeycomb ameloblastoma may be higher than unilocular ameloblastoma.
