Study On Microvascular Changes In Renal Vascular Hypertensive Rats

In order to study the microvascular changes in renal vascular hypertensive Rats,240 SD (Sprague Dawley) rats were divided into operated group including 4 weeks group,8 weeks group,12 weeks group,16 weeks group,20 weeks group,24 weeks group and corresponding control with 20 in each. Renal vascular hypertensive rats named 2K1C were established by narrowing the rats' left renal arteries using self-made metal cannulas with a caliber of 0.3mm. Before and after the operation, blood pressure of each rat was measured using self-developed non-invasive measurement system by tail volume. Then, after the operation from 4 weeks to 24 weeks as described before, kidney size and fresh weight of the two kidneys of each were measured. Dimensions of renal capsules and glomeruli were measured and number of glomeruli was counted by ink and gelatin injection method. The length density (Lv), surface density (Sv) and volume density (Vv) of the microvascular of the kidneys were measured by ink and gelatin injection and DAB method. The results were showed as follows:1. There were no significant differences of blood pressure among the control groups (P>0.05). Compared with the corresponding control, blood pressure of each operated group was significantly increased (P<0.01). In all operated groups,4 weeks group and 8 weeks groups increased significantly (P<0.01), while the others with no significant differences (P>0.05).2. Compared with the control group, the size of the left kidney of the rats in operated groups significantly diminished (P<0.01), while the right kidney significantly increased (P<0.01). There were no significant differences of the size of the left kidney between each operated group (P>0.05), neither were the right (P>0.05). In each operated group, the size of the right kidney was significantly larger than the left one (P<0.01) while no significant differences were observed in the control groups (P>0.05).3. Comparing with the control group, the weight of the left kidney in operated groups significantly diminished (P<0.01), while the right kidney significantly increased (P<0.01). There were no significant differences of the weight of the left kidney among each operated group (P>0.05), neither was the right kidney (P>0.05). In each operated group, the weight of the right kidney was significantly heavier than the left one (P<0.01) while no significant differences were observed in the control groups (P>0.05).4. Comparing with the control group, the size of left renal capsule in operated group significantly diminished. For the size of the right renal capsule, those in the 4 weeks group significantly increased (P<0.01) while those in other operated groups significantly diminished (P<0.01). In each operated group, the size of left renal capsule was significantly smaller than the right one (P<0.01) while no significant differences were observed in the control groups (P>0.05).5. Comparing with the control group, the size of left renal glomerulus in operated group significantly diminished (P<0.01). For the size of the right renal glomerulus, those in the 4 weeks group significantly increased (P<0.01) while those in the other operated groups significantly diminished (P<0.01). In each operated group, the size of left renal glomerulus was significantly smaller than the right one (P<0.01) while no significant differences were observed in the control groups (P>0.05).6. When observed under the microscope, the number of glomeruli in the left kidney in operated group became significantly less than the control group (P<0.01). For the number of glomeruli in the right kidney, those in the 4 weeks group had no significant variation (P>0.05) while those in other operated groups significantly diminished(P<0.O1) in comparison with the corresponding control. In each operated group, the number of glomeruli in the left kidney was significantly less than those in the right (P<0.01) while no significant differences were observed in the control groups.7. The rats'renal microvessel showed as the dark curve after the disposal of the ink and gelatin injection, and as the brown bead-like after the disposal of DAB. Comparing with the control group, the shape of the renal microvascular changed, the distribution of the renal microvessel was uneven with worse continuity in the operated group. 8. By illumination of ink and gelatin injection and the DAB method, it is observed that the Lv,Sv,Vv of the renal microvessel in both kidney significantly diminished (P<0.01) comparing with the control group, what's more, the density of the renal microvessel in left kidney was significantly lower than that in the right one (P<0.05) with the extension of the modeling time and the lasting function of the hypertension. Renal microvessel with the ink and gelatin injection method had a lower density than DAB method.The results suggested that injury was insulted in the renal morphology, renal capsule, glomerulus and renal microvessel of the 2K1C renal vascular hypertensive rats with the extension of the time and the hypertension, and also the renal microvascular changed, the density of the renal microvessel declined.
