| Objective: In establishing adjuvants arthritis model based on ShuXueTong injection of rats to observe the therapeutic effect of adjuvant arthritis, and the method of enzymoimmunoassay to detect the adjuvants arthritis serum TNF-a, IL-1, IL-4 expression, this paper discusses the influence mechanism.Methods: (1) The Wister female rats selected at random, 24 rat were divided into three groups: the model group, ShuXueTong treatment group﹑dexamethasone injection treatment group. Each group of rat were intracutaneously injected with 0.1 ml Sigma Freund's complete adjuvants in the right hind lamb, copy adjuvants arthritis model. (2) The medicines were given from the 12th day to the 25th day once a day to observe its effect on multiple arthritis. adjuvant arthritis was assessed: by measuring arthritic index, paw volume, general status and weight. (3) The levels of IL-1,IL-4,TNF-a were measured by ELISA. (4) The above- metioned data were analyzed with SPSS10.0statistical software.Results: (1) ShuXueTong group and Dexamethasone group could markedly improve rats general status, compared to the model group﹙P﹤0.05﹚. The increasement weight of ShuXueTong group and Dexamethasone group had markedly difference as compared to the model group﹙P﹤0.05﹚(2) There was no clearly difference of arthritis index and paw swelling in every group after modeled group. after treatment, every groups could reduce the arthritis index and paw swelling , there was difference as model group﹙P﹤0.05﹚. The arthritis index of Dexamethasone group was superior to the ShuXueTong group,but two groups had no significant difference(P>0.05).(3) The level of IL-1, TNF-a model group was higher than other treatment group. And the level of IL-4 was lower than other treatment group. After treatment, the level of IL-1, TNF-a decreased clearly in the ShuXueTong group as compared to the model group. There was significantly difference﹙P﹤0.01﹚.the level of IL-4 was recreased in the ShuXueTong group and Dexamethasone group as compared to the model group﹙P﹤0.01﹚.(4) Differences between ShuXueTong group and Dexamethasone group,indicating that the role of ShuXueTong group weaker than Dexamethasone group.Conclusion:(1) The result showed that: ShuXueTong group had a markedly effect in treating adjuvant arthritis . It could reduced the paw swelling and arthritis index, It could decrease the levels of serum IL-1, TNF-a and increase the level of IL-4. All in All ,ShuXueTong group could restrain adjuvant arthritis symptoms and suppressed cartilage and synovial tissue destruction. It is improved that the medicine has some effects on the animal model of rheumatoid arthritis.(2) The impossible mechanism was that the ShuXueTong Injection decrease inflammatory cytokines by improve the blood circulation .moreover ,it could restrain adjuvant arthritis symptoms and suppressed cartilage destruction through reduced the levels of serum IL-1, TNF-a, increased the levels of IL-4,to achieved balance of Th1/Th2. |
