[objective]As the carcinogen to mankind, the benzene have become an important occupationally dangerous factor. For effectively protecting the health of the workers occupationally exposed to benzene, by environment monitoring, occupational health inspecting and monitoring the heredity material of the workers,we understand the distributed level and characteristics of every biological indicator in workers exposed to benzene. Studied the correlationship between the biological indictors and the contact time, at the base of the examination in regular occupational disease, combine micronuclear (MN) incidence rate and chromosome damage of peripheral lymphocyte of the workers occupationally exposed to benzene, to select the more sensitive and effective early biomarkers and to provide the foundation for carrying on occupational disease prevention and cure in earlier days to the workers occupationally exposed to benzene.[methods]Using cross-sectional research methods, we conducted an epidemiological survey on the painting workshop in a large mechanical plant in Jinan.1 Research objectExposed group:The workers in paint-spray workshop with integrated occupational healthy file and good stability in a large machine production factory in Jinan. Contrast group:The manage and logistical personnel in the factory.2 Carry out occupational epidemiological investigation in work site and detect the occupational hazardous factors. Using Gilian individual air sampler and activated carbon tube to collect the air sample within continuous 8 hours, using solvent-meteorological mutually chromatogram and to calculate the concentration of the time weighted average(TWA) of the samples.3 Occupational health inspection in the crowd occupationally exposed to benzene:3.1 Blood routine:WBC, PLT, RBC and Hb3.2 Bio-chemical checks:ALT3.3 Electrocardiogram3.4 Pure oxidize-the anti-oxidize index:the super oxide Qi turns Mao (SOD)3.5 Heredities damage index:the incidence rate of micronuclear (MN) and chromosome aberration (CA) in peripheral lymphocytes.[Result]1 The results of the investigation in occupational environment:The CTWA of benzene every spray-paint workshop in investigated place are lower than the occupational exposure limits.2 Health hazards of workers exposed to benzene:2.1 The conscious symptom includes that of nervous system and irregular menstruation. The nervous system symptoms are mainly headache, giddy and decrease of memory; the rates of two symptoms in exposed group and in control group are 6.73%,3.85%and 1.61%,1.61%, the rates of exposed group are higher than that of control group. The conscious symptom occurred in the group working for 10 years. The symptom of headache and giddy occurred more earlier than decrease of memory.2.2 In the exposed group, the account of WBC and PLT are obviously lower than that of contrast group, the activities of ALT and SOD are distinctly higher than that in contrast group. The time occupationally exposed to benzene has obvious dose-response relationship with the WBC and PLT account, ALT and SOD activity. With the increasing of time exposed to benzene, the obvious differences of WBC ALT SOD activity and PLT with contrast group occur successively.2.3 The MN and CA incidence rates of every exposed group are obviously higher than those in contrast group, and has obvious dose-response relationship with the time occupationally exposed to benzene. Comparing with the indexes of blood RT, biochemistry, the obvious differences with contrast group of MN and CA incidence rates occurs in the earliest time,【conclusion】1 Exposed to benzene in low dosage for long term, damage occurs to the blood and nervous system.2 WBC, PLT, ALT, SOD, MN and CA can be the effective biomarkers of benzene occupation. The incidence rates of MN, CA and WBC are the most sensitive effective biomarkers, ALT and SOD are less than them, PLT are the last.3 The time of abnormal body appears in exposed group is after 15 years of occupational exposure to benzene. There is little significant for the abnormal rate of every index to evaluate the damage of benzene.4 It is suggested to carry out determining test of benzene heredity hurt, MN is more fit for routine detecting item in occupational healthy examination.
