Objectives:To further study the difference in hemorheology between aged rats and young rats, investigate the mechanism of Danshen injection on improving red blood cell deformability and fluidity of aged rats based on the change of hemorheology and the constituents of erythrocyte membrane. explicit the effects of Danshen injection on micro hemorheology and provide certain experiment basis for further development and clinical rational utilization of Danshen injection.Methods:Thirty wistar rats were randomly assigned to be Danshen injection high-dose group, low-dose group and model group with 10 rats in each group. Another ten younger wistar rats were acted as young control group.14 days after giving medicine,the blood are taken to measure the whole blood viscosity,plasma viscosity,hematocrit,red blood cell deformability index,and the erythrocyte membrane is taken to measure the contents of cholesterol,sialic acid,sulfhydryl and malodialdehyde, superoxide dismutase activity,Na+-K+-ATPase activity for discussing the mechanism.Results:①the whole blood viscosity and plasma viscosity of the aged rats are significantly higher (P<0.01, P<0.05) and the red cell deformability index is lower(P<0.01), than that of the young rats. Danshen injection high-dose group, low-dose group could reduce the whole blood viscosity (P<0.05)and plasma viscosity of the aged rats (P<0.05), and improve the red cell deformability index (P<0.01, P<0.05);②the content of membrane cholesterol of the aged rats is significantly higher(P<0.01) and the content of sialic acid is lower (P<0.01) than that of the young rats; Danshen injection high-dose group can obviously decrease the content of cholesterol(P<0.01) and increase the content of sialic acid in erythrocyte membrane (P<0.05);③SOD activity in erythrocyte membrane of the aged rats is lower(P<0.01) and the content of MDA is higher (P<0.01)than that of the young rats; Danshen injection high-dose group, low-dose group can improve the SOD activity(P<0.01, P<0.05) and lower the content of MDA(P<0.01, P<0.05).Cunclusions:the whole blood viscosity and plasma viscosity of the aged rats were higher than the younger rats. the deformability of erythrocyte of the aged rats was lower than the younger rats. The mechanism of that maybe have relation with the activity of the SOD and the contents of the cholesterol in erythrocyte membrane and the Sialic acid.Danshen injection can improve the aged rats'blood hemorheology through increasing the content of sialic acid and SOD and reducing the content of cholesterol in erythrocyte membrane. It indicated that the metabolism of lipid was effected, the ability of anti-oxidation was enhanced, the content of oxygen radical was decreased and the carrying capacity of negative charge in erythrocyte membrane was improved.
