Objective:Investigated for the treatment of cerebral infarction and other ischemic encephalopathy whether XNJI also be used for treatment of cerebral ischemia induced lupus encephalopathy (CNS-SLE), and to analyze its mechanism.Methods:SLE model mice were divided into Model group, treatment group XNJI, prednisone and cyclophosphamide treatment group; 4 months for drug intervention cranial MRI scans MRI;Analysis of brain white matter deep cerebellar hemispheres, basal ganglia and brainstem abnormalities, and cerebrospinal fluid levels; 4 months of drug intervention step down method using CNS-SLE determination of learning and memory in mice; Interim and final collection of drug intervention serum, enzyme-linked immunosorbent assay of serum cytokines TNF-α, IFN-γ; 4 months after drug intervention, taking CNS-SLE pathological analysis of brain tissue in mice, immunohistochemical analysis of VCAM-1, Fas/FasL protein, P38 protein expression and phosphorylation; By Real time RT-PCR (SYBR dye) assay in brain tissue of vascular cell adhesion molecule-1 (VCAM-1) mRNA expression.Results:4 months after drug intervention of Chinese medicine and western medicine treatment group relative to the model group the symptoms of the limbs was relatively mild erythema; Map based on MRI analysis, we found that, Type groups exist abnormal signal lesions and brain hemorrhage, while Chinese medicine and western medicine group lessened a lot, Cerebrospinal fluid content of the three groups fairly, no special exception; In step down test, the learning ability of rats three reaction time and number of errors was no significant difference in memory retention test results showed that compared with the model group, treatment group, the latency of jump significantly increased (P<0.05), the number of errors without Significant difference (P> 0.05); Serum IFN-γ, TNF-αat the same time the serum levels of three groups no significant difference (P> 0.05), but IFN-γand western medicine group at the end of the model group were significantly lower than serum levels of the mid (P<0.05). Model of pathological brain tissue image shows a large number of inflammatory factors, there is bleeding, there is edema, neuronal cell proliferation in general; Chinese medicine and western medicine such pathological features greatly reduce or disappear, neurons are active hyperplasia; Immunohistochemistry showed that VCAM-1 and phosphorylation of P38 in Chinese medicine and western medicine in brain tissue was significantly lower than model group (P<0.05); FAS, FASL, P38 in the three groups of brain tissue levels were not significantly different (P> 0.05); three groups of mice brain tissue expression of VCAM-1 mRNA levels were not significantly different. Conclusion:Xingnaojing intervention improved the SLE model type mice brain hemorrhage, inflammation, edema and other symptoms, enhance memory in mice; Lowered the VCAM-1 in brain tissue and phosphorylation of P38 protein expression, thus affecting the SLE mouse model of lupus encephalopathy secondary to the process.
