Objective 1. To observe the inhibition role of growth on SGC7901 cells acted by Weikang granules (WK).2. To investigate the potential mechanisms of the inhibition role of growth on SGC7901 cells acted by Weikang granules (WK).3. To investigate the effects of Weikang granules (WK) for the growth of gastric cancer in nude mice impanted with gastric cancer cell SGC7901.Methods 1. In vitro,the effect of Weikang granules (WK) on SGC7901 cell proliferation was analyzed by MTT assay.2. Detected the expression of different dosage of Weikang granules on the impact of Bcl-2,Bax,Mcl-1,Bcl-XL,P53,PARP and LC3 protein.3. In vivo,50 nude mice, which had been inoculated hunman gastric cell line SGC7901 subcutaneously, and after sixteen days, they were divided into five groups at random and received treatments every day. The five groups were:control group, fluorouracil group (5-FU group), low-dosage,medium-dosage and high-dosage Weikang granules groups(2.4,4.8,9.6g/kg). We measured the volume of transplanted tumors every three days. After 25 days, the mice were killed and the tumors were immediatedly removed for further studies. We calculated the volume and weight of tumors.Results 1. In certain range of concentration, Weikang granules (WK) could obviously inhibite the-growth of SGC7901 cells in dose and time dependent manner.2. The expression of Bcl-2,Bax,Mcl-1,Bcl-XL,P53,PARP protein in different dosage groups were not changed obviously, but the expression of LC3-I protein was decreased, while the expression of LC3-Ⅱprotein was increased, in dose and time dependent manner.3.25 days later, the rate of growth inhibition of experimental groups and 5-FU group were apparent compared to the control group. What's more, the rate of growth inhibition of medium-dosage and high-dosage Weikang granules groups were different with statistical significance(P<0.05).Conclusions 1.Weikang granules (WK) could obviously inhibite the growth of SGC7901 cells in dose and time dependent manner.2. Weikang granules (WK) may inhibite the proliferation of SGC7901 cells via autophagy.3. Weikang granules (WK) could inhibit the growth of cancer cell in vivo.
