Exploration Of Mechanism Of The Restriction Of Rats' Endometriosis Recurrence By The Herbal Medicine Of HuoXue Xiao Yi Throuth The "AAA" Process

ObjectiveWe explore the mechanism of the restriction of recurrence of endometriosis (EMs) using the herbal medicine of HuoXueXiaoYi through observing the changes of the ectopic endometrium volume, the pathologic, and the expression levels of ICAM-1,MMP-9,VEGF.MethodsGroup:Seventy SD female rats were randomly divided into seven groups and every group has ten rats. There are one normal control group, two model control groups, two gestrinone groups, and two HuoXueXiaoYi groups. Except the ten in the normal control group, the rest sixty rats were made by autotransplantation of their own endometrium.Dose:the rats started to be administered medicine in the next day after operations. For the HuoXueXiaoYi groups, the rats were administered 1 ml/100g medicine with the concentration of 1250mg/ml once per day, i.e.12.5g/kg/d which corresponds to 2g/kg/d for an adult. The gestrinone groups were administered 1 ml/100 g medicine with the concentration of 0.026mg/ml twice per week and administered salt water in the other five days, i.e.0.26mg/kg/d which corresponds to 0.042mg/kg/d for an adult. For the normal control one as well as the model control ones, the rats were administered salt water once per day. The medicine was injected into the stomach directly. After being administered for three weeks, the normal control group, one model control group, one gestrinone group, and one HuoXueXiaoYi group were exsanguinated and their ectopic endometriums were cut with narcosis and then killed. The rest thirty rats were performed in the same way in the next three weeks after stopping administering.Observation:the changes of the ectopic endometrium volume, the pathologic change, and the changes of the expression levels of MMP-9,ICAM-1,VEGF.Statistics:SPSS (v 15.0) was been applied to perform the statistics of data obtained from experiments. Every value shown in this thesis was taken the form that the average±standard deviation. The difference standard between two group of data with statistic meaning is P<0.05.Results.1 The difference of the entopic endometrial volumes has statistic meaning (P<0.05) between the HuoXueXiaoYi group and the model control group, for either the case of being performed as soon as the administering was stopped (CaseⅠ) or the case of being performed in the next three weeks after administering was stopped (CaseⅡ).2 In ectopic endometriums, the expression levels of ICAM-1 of the HuoXueXiaoYi group are lower than either the model control group or the gestrinone group for Case I and their difference has statistic meaning (P<0.05). For CaseⅡ, the expression levels of ICAM-1 of the HuoXueXiaoYi group are lower than the model control one and their difference has statistic meaning (P<0.05). In blood serum, the expression levels of ICAM-1 of the HuoXueXiaoYi group are lower than the model control group for either Case I or Case II and their difference has statistic meaning (P<0.05).3 In ectopic endometriums, the expression levels of MMP-9 of the HuoXueXiaoYi group are lower than either the model control group or the gestrinone group for Case I and their difference has statistic meaning (P<0.05). For CaseⅡ, the expression levels of MMP-9 of the HuoXueXiaoYi group are lower than the model control one and their difference has statistic meaning (P<0.05). In blood serum, the expression levels of MMP-9 of the HuoXueXiaoYi group are lower than the model control group for Case I and their difference has statistic meaning (P<0.05).4 In ectopic endometriums, the expression levels of VEGF of the HuoXueXiaoYi group are lower than either the model control group or the gestrinone group for Case I and their difference has statistic meaning (P<0.05). For CaseⅡ, the expression levels of VEGF of the HuoXueXiaoYi group are lower than the model control one and their difference has statistic meaning (P<0.05). In blood serum, the expression levels of VEGF of the HuoXueXiaoYi group are lower than the model control group for Case I and their difference has statistic meaning (P<0.05). Conclusions1 The approach is feasible that the rat model of EMs and administering in the next day after operations to simulate the environment of recurrence of EMs.2 The herbal medicine of HuoXueXiaoYi can cause the decrease, atrophy, and even disappearance of the ectopic endometrium.3 The herbal medicine of HuoXueXiaoYi can restrain the adhesion and aggression of ectopic endometriums. It can also restrain the formation of local blood vessel to reduce the ability of planting, aggression, transfer of ectopic endometriums and sequently restrain the formation and development of EMs.4 The current experiment proved that there is a strong dependence of recurrence of EMs on the "AAA" process. By restraining expression of some factors of the "AAA" process, the herbal medicine of HuoXueXiaoYi can prevent recurrence of EMs and these factors can be restrained for a long time.5 The herbal medicine of HuoXueXiaoYi has advantages over gestrinone to prevent recurrence of EMs.6 The current study showed "the preventive treatment of disease" in the traditional Chinese medicine.7 The current experiment investigated the dependence of EMs recurrence on the factors, such as ICAM-1, MMP-9, VEGF, etc., and the short-period and long-period impact of the herbal medicine of HuoXueXiaoYi on these factors. However, the experiment cannot give a systematical and complete explanation for the recurrence mechanism of EMs and the mechanism of interactions of it with the herbal medicine of HuoXueXiaoYi. These will be our father investigation objectives.
