Objective:By setting zhe rat model of IRI,we try to anylsis significance of VEGF and HIF-la in lung ischemia-reperfusion injury.Also compare profection of Ambroxol.Methord:To set zhe rat model of IRI,we occluded the left pulmonary hilum for 60 mins and reperfuse 6 hours.50 healthy femal rats were randomly devided into 5 groups.①Control Group:Ras were taken directly after anesthesia and thoracic incision,without assist of respirator.This group was served controls to other groups.②IR group:with the assist of incision of trachea and respirator,we sternotomied the left lung hilm and rendered ischemia by ligating the pulmonary hilum for 60 minutes followed 6 hours.After executing the rats,we get the specimen.③IR-Sham Group: Without ligating the pulmonary hilum, we get the the specimen.④ⅠGroup: Ligating the pulmonary hilum for 60 minutes,we didnn't open the clamp,and got the pulmonary specimen⑤Ⅰ-Sham Group:After incision of trachea and respirator, We didnn't ligate the left pulmonary hilum and got the pulmonary.Every group was divided 10 ras.we shoulg give other 10 ras,if the rats of group was died.After finishing the experiment,we measured W/D, IL-1β,TNF-α.We investigated Pathologieal changes of lung by Pathology, IHC and imaging analise,expression of VEGF and HIF-la.Then, in order to estimate the ptotection of Ambroxol,we devided 4 groups with 40 healthy femal rats randomly.(1)Control Group:without any treatment,we sternotomied and got the lung tissure,also got the blood from left atrium.This group was comparision of other groups. (2)IR Group:we sternotomied and open the left lung hilm after ischemia by ligating the pulmonary hilum for 60 minutes with microclamp.Nacl 0.9% was administered intravenously through femoral vein since ligating the pulmonary hilum.(5ml/kg/h).(3)IR-Sham Group:we sternotomied and dissociate the pulmonary hilum,without ligating the pulmonary. Nacl 0.9% was administered intravenously through femoral vein since suturing thorax. (5ml/kg/h). (4) Ambroxol Group:we sternotomied and open the left lung hilm after ischemia by ligating the pulmonary hilum for 60 minutes with microclamp.Ambroxol was administered intravenously through femoral vein.(5ml/kg/h).Rats ware executed after 6 hours. After finishing the experiment,we measured W/D, IL-1β,TNF-αand MPO.We investigated Pathologieal changes of lung by Pathology, IHC and imaging analise,expression of VEGF and HIF-la.Result:Wet/Dry Weight Ratio:There is statistical significance between IR Group and Control Group,IR-Sham Group,ⅠGroup,Ⅰ-Sham Group.(P<0.05),and amout of IL-1β,TNF-αincreasing also, Comparing with Control Group,IR-Sham Group,ⅠGroup,Ⅰ-Sham Group.But,there is no statistical significance betweenⅠGroup andⅠ-Sham;Lung edma in IR group moreheavily than other 4 Groups. Comparing with Control Group,,ⅠGroup,Ⅰ-Sham Group.There is statistical significance between Groups in expression of HIF-1 a. (P<0.05).But no statistical significance between IR Group,Ⅰ-Sham Group andⅠgroup. There is statistical significance between Groups in the expression of VEGF and there is statistical significance between IR-Group andⅠgroup,Ⅰ-Sham Group. (P<0.05).but no statistical significance compareing whith control group,IR-Sham group. Correlation analysis in expression of VEGF and HIF-la:the expression of VEGF and HIF-1a is correlated with Pearson correlated analysis.so there is correlated in the expression of VEGF and HIF-la in the LIRI.Ambroxol Group:amount of IL-1β, TNF-αand MPO are lesser than IR Group with no Ambroxol.W/D also lesser and PaO2 increasing.With the light micro,we acquire lung edam and hemorrhage in lung are lessen. Congratulation:HIF-1a and VEGF participate the LIRI and Ambroxol has the effect of lung protection.
