In our country, the morbidity of PCa has rised to the third place in male genitourinary malignant tumor system, PCa has already affected our men's quality of life and life expectancy who is over the age of 50, and has become a more and more important topic in the urology field. Androgen deprivation therapy, molecular immunization, radiotherapy and chemotherapy applied for patients with advanced or metastatic PCa has made certain curative effect. In recent years, antiangiogenesis of molecular target therapy has shown more and more good prospect. Research has proved, vascular endothelial growth factor (VEGF) is highly expressed in PCa. In the recent ten years, more and more authors proved that macrophage migration inhibitory factor (MIF) and tumorigenesis are closely related, in addition to its proinflammatory behavior, also can promote angiogenesis to promote the tumor formation. Our experiment is aim to discuss the significance of MIF, VEGF and MVD (marked by CD34) in carcinogenesis, progression, angiogenesis and their relationship of PCa, and provide a new approach for the diagnosis and treatment of PCa.Materials and methods:A total of 63 PCa and 20 BPH patients from department of urology, with accession from January 1995 to October 2008, were selected in this study. The median age was 69.67±8.35 years and 71.20±6.17 years respectively in PCa and benign prostatic hyperplasia (BPH) patients, and the statistics shows no significant difference (P<0.05). Gleason score 2-6 score was in 13 cases,7 score was in 17cases,8-10 score was in 33 cases; AJCC TNM clinical stage:Ⅱstage was in 24 cases,Ⅲstage was in 25 cases,Ⅳstage was in 14 cases. Immunohistochemistry (EliVision method) was used to detect the expression of MIF, VEGF and MVD in 63 PCa and 20 BPH cases. All dyeing slice was assessed by two pathological doctors in a double-blind method. The score criteria of MIF and VEGF apply the method of Liangzhong Xu et al. MVD count criteria using Weidner's method. SPSS 16.0 statistical software was applied to anlysis the data and statistical significance was defined as P values less than 0.05.Results:(1) In BPH and PCa, the positive rates of MIF was on the rise from 15.00% to 82.54% respectively, showing significant difference (P<0.05); The expression of VEGF was also on the rise, from 35.00% to 84.13% respectively, showing significant difference (P<0.05); the amount of MVD was also on the rise, from 35.70±6.18 to 64.35±17.41 respectively, showing significant difference (P<0.05). (2) In Gleason score 2-6 score,7 score and 8-10 score, the positive rates of MIF was 53.85%,82.35%and 93.94%respectively, showing significant difference (P< 0.05);In clinicalⅡ,ⅢandⅣstage, the positive rates of MIF was 66.67%,88.00% and 100.00% respectively, showing significant difference (P<0.05). The positive rates of VEGF was 61.54%,82.35% and 93.94% respectively in three Gleason score group, showing significant difference (P<0.05); and the positive rates of VEGF was 75.00%,84.00% and 100.00% respectively in three clinical stage group, showing significant difference (P<0.05). The amount of MVD was 47.23±5.04,55.06±7.06 and 81.61±9.93 respectively in three Gleason score group, showing significant difference (P<0.05); the amount of MVD was 54.83±13.18,72.00±15.70 and 80.50±12.89 respectively in three clinical stage group, showing significant difference (P<0.05).(3)The analysis of the expression of MIF, VEGF and MVD in PCa shows, there were positively significant correlation between MIF and VEGF, MIF and MVD, VEGF and MVD (P<0.05)Conclusion:(1) The expression of MIF, VEGF were more increased in Pca than BPH, and the amount of MVD was more increased in Pca than BPH.(2) With the rise of clinical stage and Gleason score grading, MIF and VEGF positive expression, MVD amount were significantly increased.(3) Expression of MIF, VEGF and MVD were positively correlated in PCa tissues(4) Through this experiment, we can enhance the understanding of PCa vascular regulatory mechanisms, combined detection of MIF, VEGF and MVD can provide a reference value for PCa occurrence development and prognosis. | | Keywords/Search Tags: | Prostate tumor, MIF, VEGF, MVD, CD34, Immunohistochemistry | PDF Full Text Request | Related items |
