Arsenic compound is a classical carcinogens, which has DNA damage effect on organism. Roxarsone not only has significantly effect on the DNA damage, but also has potential genetic toxicity to organism in the environment although the toxicity of roxarsone is lower than inorganic arsenic. In this paper, single-cell gel electrophoresis was used to demonstrate the roxarsone's side-effect on mice liver and lung cells DNA, and may lay basic foundation of the roxarsone to the genetoxicity research and risk evaluation to organism in the environment.The ICR mice were exposed to different concentration of roxarsone(1/2LD50, 1/5LD50,1/10LD50, and control group), which were exposure time to 1,2,4,6,8d respectively. Then, the mice were sacrificed after 1,2,4,6,8d, the liver and lung of mice were made into single cell suspension, cell density was adjusted in the 106~107/ml. Based on the modified alkaline single-cell gel electrophoresis, the content of tail DNA, tail moment and olive tail moment were measured by the CASP software. The results showed that the liver and lung cells of mice were exposed to the dose of 1/2LD50, 1/5LD50, which could induced DNA damage. To compared with the control group, the damage caused by roxarsone was obviously great and the differences was significant (P<0.05), and the exent of DNA damage showed a striking dose-response relationship and time-response relationship.At the same time, the liver and lung cells of mice also showed DNA damage at the dose of 1/10LD50, which have significant difference compared with the control group (P>0.05).Results showed that roxarsone can caused the DNA breakage and migration seriously, and have significant difference compared with the control group.Immunohistochemistry test have good localization to VEGF and CD34 expression on mice liver and kidney tissue which induced by roxarsone, VEGF and CD34 have significant expression at the dose of 1/2LD50, It is positive that VEGF was stained brown on the cytoplasm, CD34 was localized in the cytoplasm or cell membrane of vascular endothelium. At the dose of 1/5LD50,1/10LD50 can not be detected. The single-cell gel electrophoresis and immunohistochemistry test were showed that roxarsone has DNA damage to the mice cells,and the former could detect earlier cell DNA damage. The single-cell gel electrophoresis could be used as an effective method to detect the DNA damage of organoarsenic compound in the environment.
