Effect Of Cisplatin On IL-18 Contents In Rats With Endometriosis By Peritoneal Lavage

Objective:By observing effect of Cisplatin on rats with endometriosis (EM) of the levels of IL-18 in Abdominal Cavity Fluid,we investigate the mechanism of Cisplatin on rats with endometriosis.Methods:We set up endometriosis model by rat self-endometria transplantation. By Checking forty-five SD rats'vaginal smears,we select rat with estrous cycle of four-five days and two consecutive normal estrous cycle as experimental subjects.we set up endometriosis model in the third estrous cycle. Before seting up endometriosis model, nine rats served as sham operation group (A), the remaining rats were modeling. Sham group of rats only stretch the uterus by clamp after laparotomy,the other dealing is the same as surgery group. After cut the left side of the uterus into small pieces 1～2mm, we planted them in adipose tissue around the right ovary, mesenteric, subcutaneous and pelvic walls on both sides.we send the remaining tissue for pathological examination To confirm that the graft is uterine tissue.30 of 36 rats were confirmed successful model by pathology.Model rats were randomly divided into 3 groups. Model control group (D) contains 10 rats,cisplatin group (B) contains 9 rats and saline group (C) contains 11 rats. Group D were not given any treatment while group B and C were douched with Cisplatin injection of 20mg/kg and normal saline of 10ml in rats'abdominal cavity. After four weeks, the rats of four groups again receive laparotomy.We observe the growth of ectopic endometrium,wash the abdominal cavity by normal saline of 2 mL～3mL,and collect peritoneal fluid. The levels of IL-18 in Abdominal Cavity Fluid were measured by ELISA.Results:Gross specimen observation:four weeks after administration, the rats of four groups receive re-laparotomy to observe the growth of ectopic endometrium, the Lesions of normal saline and model control group increased over the previous volume, and the Lesions of cisplatin group decreased compared with the previous volume.IL-18 concentration:(1) After the models were made,the levels of IL-18 in Abdominal Cavity Fluid were significantly higher compared with those before the models were made (P<0.05). (2)The levels of IL-18 in normal saline group and model group were significantly higher compared with those in non-surgery group (P<0.05). (3) Compared with model group, The levels of IL-18 in Cisplatin group were significantly lower. There was significant difference of IL-18 between two groups(P<0.05).Conclusion: Cisplatin can obviously improve the abdominal microcirculation and its treatment mechanism may probably operate by decreasing the level of IL-18 cell agent to inhibit the production of the endometriosis.
