Objective:Purpose of this study is that explore the effect of osteoinductive activity ofβ-tricalcium phosphate/P-15 peptide (P-TCP/P-15) composite materials and was to give a preliminary experimental evidence and theoretical basis as bone graft materials.Methods:1.β-TCP was put into P-15 peptide solution,Incubation was carried out at room temperature for 24 hours with gentle shaking to ensure the peptide full absorption intoβ-TCP, unadsorbed peptide was removed from theβ-TCP by PBS, Theβ-TCP/P-15 composite was dried and then stored at room temperature in moisture-proof containers;2.C2C12 cells were cultured on the surface ofβ-TCP/P-15 andβ-TCP for 6 days, samples were obtained 2,4,6 days after culture, the morphology of C2C12 cells were observed under scanning electron microscopy; Cell proliferation was detected by CCK-8 method; Total cell protein was examined by BCA method; Cell alkaline phosphatase(ALP) activities were assayed by ALP kit.3.Bone defected were conducted in mandubular body. The both materials of experimental group 1 (β-TCP/P-15)and experimental group 2 (β-TCP) were implanted in the same rabbits on the left or right mandibular defect at randomly, control group was randomly selected the left or right mandibular defect,which no material was implanted, animals were sacrificed at 4,8 and 12 weeks after surgery, Evaluate the grade of repair by gross observation, histological staining and determination of the new bone area.Results:1. Both cell proliferation and total amount of protein of C2C12 cells ofβ-TCP group andβ-TCP/P-15 group increased with time, The cell proliferation and total amount of protein of C2C12 adhered top-TCP/P-15 were obviously higher than that of C2C12 adhered toβ-TCP (P<0.05). After 2 days culture ALP activity of C2C12 of two groups were not detected. After 4 days and 6 days of culture, ALP activity of C2C12 of two groups increased with time, ALP activity of C2C12 adhered toβ-TCP/P-15 were obviously higher than that of C2C12 adhered toβ-TCP. (P<0.05).2.The evaluation of gross observation and histological staining ofβ-TCP/P-15 group are better than that ofβ-TCP group at each point; Both new bone area ofβ-TCP group andβ-TCP/P-15 group increased with time, there was significant difference (P <0.05) in the same group at different time,β-TCP/P-15 formed bone area increased rapidly; each time pointβ-TCP/P-15 formed bone area was stronger than the control group (P<0.05).Conclusion:β-TCP/P-15 composite has good biocompatibility, bone conduction, degradation and bone inducing activity, and can promote the repair of bone defects. Which was expected as a material for bone repair treatment of mandibular defects.
