Objective:Established a mouse model of herpes neuralgia and gave feeding cimetidine treatment, through analysing the changes of regulatory T lymphocyte subgroup in the pathogenesis of herpes neuralgia, as well as observating the reaction and intervention of cimetidine on T lymphocyte subgroup during treatment, explored the possible mechanism of therapeutic.Methods:Prepared herpes group (hsv group), herpes cimetidine treatment group (hsv / cmd group) and contrastive group (NS) animal model respectively .Collect anticogulant outside from all mouses in the treatment after 8 days, detected quantity of T Lymphocyte subgroup by flow cytometry, included the numbers of CD4+ T cells and CD4+ CD25+ regulatory T cells, and percentage of CD4+ / CD8+ T cells,and percentage of CD4+ CD25+regulatory T cells accounted for CD4+ T cells.Results:①In peripheral blood,the numbers of CD4+ T cells and CD4+CD25+ regulatory T cells, and percentage of CD4+/CD8+ T cells, and percentage of CD4+CD25+regulatory T cells accounted for CD4+ T cells of mouse which in herpes group were observably lower (p <0.05),the numbers of CD4+ T cells and CD4+ CD25+regulatory T cells, and percentage of CD4+/CD8+ T cells and percentage of CD4+CD25+ regulatory T cells accounted for CD4+ T cells were observably higher (p <0.05) after the treatment of feeding cimetidine.②There was few difference(p> 0.05) with numbers of CD4+ T cells and CD4+ CD25+ regulatory T cells, and percentage of CD4+ / CD8+ T cells, and percentage of CD4+CD25+ regulatory T cells accounted for CD4+ T cells in the treatment group and contrastive group.Conclusions:①Depletion in numbers of CD4+ T cells and CD4+ CD25+ regulatory T cells of the mouse infected with herpes resulted in attenuation of systemic and part catastaltic response to inflammation, that may be one of the reasons which brought on herpes zoster neuralgia.②Cimetidine could enhance the immune suppression function and play a therapeutic effect by increasing the numbers of CD4+ T cells and CD4+ CD25+ regulatory T cells.
