Tomato is a common vegetable in our daily life and has high nutritional value. The majordifficulties in tomato breeding are the effects of plant diseases and insect pests andrequirement of people for high quality of tomato. Compared to the traditional breedingmethods and transgenic technology, ion beam mutation breeding has high mutation rate,stable characters and other merits and so is widely used in study of crop breed improvement.In this study, the effects of ion beam and soybean DNA mediated by ion beam on the geneticmaterials of tomato and its expression in tomato were discussed with cytology, RAPDmolecular markers and SDS-PAGE in order to provide basis for ion beam mutation breedingof tomato. The main results are as follows:1.Compared to controls, N+and Ar~+ion have obvious effects upon the cell division indexand chromosome aberration rate. Within the scope of1-4×1017ions/cm2,, the cell divisionindex declines and chromosome aberration rate increases as the dose of ion beam adds.Meanwhile, various kinds of chromosome aberration happen in one cell because of the highdoses of ion beam. Under the same dose conditions, N+ion has greater influence on celldivision index and chromosome aberration rate than that of Ar~+ion, and its suppression to celldivision is stronger.2. The results of RAPD study show that genom DNA of tomato is affected by N+ion, Ar~+ion and soybean DNA injected by N+and Ar~+ion when compared with contrast. As injectedwith the same dose ion beam, the aberration rate of tomato genom DNA influenced by Ar~+ionis greater than that by N+ion, and DNA fragment has more effects on tomato genom DNAthan those of full-length DNA.3. As is indicated by results of protein SDS-PAGE spectrogram, compared with controlgroup, there are some changes in protein spectrogram of tomato injected with N+ion, Ar~+ionand oybean DNA mediated with N+and Ar~+ion. The changes caused by ion implantation aremain in the reinforcement or weakening and decrease or increase of some protein bands,while changes appear decrease, increase, weakening or reinforcement of some protein bandsin protein spectrogram of tomato treated by soybean DNA which was mediated into tomatowith ion.
