Porcine Contagious Pleuropneumonia is contagious respiratory disease caused by Actinobacillus pleuropneumoniae (App) infection. Antibiotic treatment easyly produces drug resistance. So it is important to find new drug targets and design new drugs to this pathogen.Methods:1?Based on bacterial cell division related gene sequences of GenBank, distribution of app cell division related genes in different App sertypes was determined. Acording to sequences of App ftsZ gene family in GenBank, primer pare of these genes was designed. ftsZ gene family were amplified by PCR and sequenced.2\Acording to App ftsZ gene sequences in GenBank, primer pare of App ftsZ gene was designed. App FtsZ gene was amplified by PCR,sequenced and analysized by bioinformatics. Recombint plasmid pET28-FtsZ was constructed. Rcombinant protein rFtsZ was expressed and purfied, then immunized mice. Antiseria to rFtsZ was obtained. FtsZ expression in App was detected by West-blot.Results:21 cell division-related genes of Actinobacillus pleuropneumoniae were detected. Most of these genes exist in various serotypes, and few genes are different in different serotypes.12 Fts gene had been cloned from App ser 3. Of these, ftsZ gene was sequenced and analysized by bioinformatics. App ftsZ gene was 1212 bp length, containing an open reading frame (ORF) with the start codon ATG and termination codon TAA, encoding a protein of 403 amino acids. The theoretical molecular weight of this protein is 42.6kD, its isoelectric point is 4.72. And similarity is 96% between App serum type 1, type 4, type 5, type 8, type 10 and type 11. Phylogenetic analysis showed that ftsZ gene of different serotypes App belonged to the same branch in evolution. And protein sequence analysis showed that FtsZ protein is high homology in different serotypes of APP.Conclusions:this study layed a foundation for further study on molecular and cellular mechanisms and ftsZ gene roles in App cell division. FtsZ may become a target of general medicine.
