According to the need of the test to change the starting time of superovulation and thetime of ovulation, fertilization and embryonic development of mouse, in order to take asufficient number of mature oocytes, zygotes and early embryos, to content the needs ofresearch workers on oocytes. zygotes and early embryos at different development stages.1.Choose the female Kunming mice, Using vulva state observation, vaginal smears todetermine the stage of the estrus cycle. Then make ovarian tissue sections to observe folliclegrowth dynamics during estrus cycle. On the basis, superovulate mice in each estrus stagerespectively, and track the result of follicular development and embryos collection, determinethe suitable stage of the estrus cycle for superovulation.The result is that during the estruscycle, the vulva status, vaginal smears and follicle growth dynamics are connected with eachother. The result of superovulation of mouse in proestrus was significantly better than others.2. Using immunohistochemistry to study the distribution and expression pattern ofFSHR, of LHR, EGFR, IGF-I R in mouse ovaries during the whole estrus cycle. Choose themice in dioestrus and proestrus, then inject them with EGF, the dose is0ng/g,5ng/g,10ng/gand20ng/g. Study the status of follicular development and embryo collected results aftersuperovulation. The results is that FSH and LH played an important role in antral follicle,while EGF and IGF-Ⅰplayed an important role in preantral follicle. The EGF dose of5ng/g,10ng/g,20ng/g could promote the effect of superovulation on mice in dioestrus andproestrus.3. Superovulate the Kunming mouse in proestrus, inject PMSG respectively in12:00,16:00,20:00. Study the time of ovulation, fertilization and early embryo development.The result is that superovulating mouse at the starting time of16:00, the curve of ovulation,fertilization and embryo development is relatively stable, the effect of superovulation is ideal.Beginning at20:00, the development of embryos was stable and was3~4hours later than16:00group. Superovulating mouse at12:00, the development of embryos was3~4earlierthan16:00group, but the effect of superovulation was not suitable. In summary, vulva state observation and vaginal smears can be used for judgment of themouse estrus cycle stage, the effect of superovulation of mouse in proestrus is best. FSH andLH mainly regulate the development of antral follicles, and EGF,IGF-Ⅰmainly regulate thedevelopment of preantral follicles. Superovulate mice in different starting time, the time ofovulation, fertilization and early embryo development had apparent regularity. It provide areference on mouse embryonic process.
