Leaf is one of the most important parts for the plants, and also is the main places of photosynthesis synthetic organics. Leaf type is the significant agronomic characters of the rice and other cereal crops, related to the high yield and yield stability and, therefore, has been the focus of attention of the rice genetics and breeding scientists and cultivation experts. We found a leaf type mutant-Rumpled and Twisted leaf2(rtl2) in the mutant library of our laboratory. Revertant of eg] is mutagenized by methyl sulfonic acid ethyl ester (EMS). This paper contains the morphological identification, genetic analysis and electron microscope observation etc, and purpose gene location through the map-based cloning method. This laid a foundation for elucidating the molecular mechanism.The experimental materials of my paper, rtl2mutant has the similar phenotypes with rtl1mutant. The base of the leaves also exhibited a rumpled and twisted phenotype. which is the most significant feature of the tll1mutant. During new leaf elongation, the twisted leaf prevents the blade tip extending from the sheath, thus forming an'O' shape. With the growth of new leaves, the previously malformed leaves will eventually extend, but retain the rumpled and twisted phenotype. But after heading, the panicle of rtl2mutant also twisted seriously. In addition, compared with wild type, there is no change significant. Through the map-based cloning, we eventually locate rtl2in chr.8. By using electron microscopy to scan the surface of the leaf, we found that the texture of the wild type is very beautiful, however, the texture of the mutant is also twisted.
