Fritillariaunibracteata Kisao et K.C.Hisa is an important traditional Chinese herbal medicine,often used for Clearing heat and depressing coughs,expectorant.Due to natural Fritillariaunibracteata Kisao et K.C.Hisa mainly grow higher attitude region ,longer growth period,lacking of resources.so it exists long term inadequate supply. The study establish valid Micropropagation system of Fritillariaunibracteata Kisao et K.C.Hisa by tissue culture. their bulbs were used as explants in the medium of MS. Through different kinds of hormone and propotions to choose best medium for adventitious buds ,callus induction , regeneration induction of bulblet growth ,rooting and bulblet formation.This paper focuses on primary application and the initial research work, thus provided the evidence on the feasibility of industrialized production. The results are shown as follows:1 Tender stem segments was optimal for explant Primary culture.The best sterilizing method for tender stems was disinfected with2 Through researehing the effect of different hormone combination on adventitious buds,callus induction and regeneration induction of bulblet growth,we found that:(1)The differentiation and induction for adventitious buds,the optimal medium was MS+NAA 2.0mg/L+6-BA 0.5mg/L(2)The optimal medium for callus induction was MS+2,4-D 1.0mg/L+NAA 0.5mg/L(3)The optimal medium for regeneration induction of bulblet was MS+NAA 1.0mg/L+6-BA 0.5mg/L3.Stage of multiplication and differentiationDuring the Stage of multiplication and differentiation, for bulblet,it was found that MS medium supplemented with MS+NAA0.5mg/L+6-BA0.5mg/L was suitable.4.Stage of rooting and bulblet formationDuring the Stage of rooting and bulblet formation, the result showed that the category and the concentrations of hormone and sugar affeeted rooting and forming of bulblets.The NAA benefited the rooting ,the better concentration was the sugar benefited the bulblet formation, the better concentration was.the better rooting and bulblet formation medium was 1/2MS medium supplemented with sugar and NAA2.0mg/L5.Stage of transplantationThe stages in transplantation ,that was sand bed preparing , domestieation, PseudoPlantation and transplant soil.the survival rate of transplantation could be 90%.
