Stratification Time And Gibberellic Acid Concentration On Smuggling Effects Of Tissue Culture

Syringa reticulate var.mandshurica is a deciduous small tree of the genus Syringa in the family Oleaceae.It is widely distributed in many industries and plays an important role.In order to improve the quality of tissue culture seedlings and large-scale seedling cultivation techniques of Syringa mandshurica,we used the seeds of Syringa mandshurica as materials to study the germination of sterile seeds,the induction of clustered buds and the occurrence of adventitious buds through different stratification time and gibberellin concentration treatments.The results are as follows:Sterile seed germination and cluster bud induction experiments were carried out after different explants sterilization treatments with different stratification time of S.lilac seeds as materials.The results showed that the sterilization and inoculation methods with 5%Na Cl O+extrusion embryo had the lowest bacterial infection rate of 8%.In MS+6-BA 5 mg/L medium,the seed germination rate and cluster bud induction of three weeks stratification were the highest,reaching 100%.After two weeks of stratification,the proliferation rate of clustered shoots was the highest,300%;arbuscular buds were induced by seed germination at different stratification time.The highest induction of arbuscular buds was induced by seed germination at 5 weeks,reaching 2.6 cm after 3 weeks.Germination of sterile seeds and induction of clustered shoots were studied using seeds of S.mandshurica treated with different concentrations of gibberellin as explants.The results showed that gibberellin added to the medium had a significant indigenous effect on seed germination.For the seeds without stratification,the embryo was not extruded,and the germination rate was up to 100%when GA30.05 mg/L was added to the germination medium.Adding GA3to the forward medium of cluster bud induction was helpful to plant elongation.When the concentration was 0.5 mg/L,the average elongation could reach 10mm,and the maximum per plant could reach 33 mm.The elongation experiments of clustered shoots were carried out by soaking seedlings with GA3and adding GA3to the medium.The results showed that there was no elongation effect on clustered shoots,and the clustered shoots were browning.
