Cell apoptosis is a process of cell death which is resulted from the death program that triggered by outside and inside factors. It's an important mechanism for multicellular biology to maintain self stabilization. The essence of ovarian atresia is the apoptosis of oocyte and granule cell.The transcription factor Foxo1 has an important role in development,growth,metabolism,cell cycle and apoptosis through transcriptional regulation and signal transduction pathways. To investigate the effect of Foxol on apoptosis of mice granule cells and explore the molecular mechanism of ovary atresia, the experiment constructed an enkaryotic expression vector FOXO1 to overexpress FOXO1 in mice granule cells.The results indicated that:①in the normal situation, mice granule cells express Foxo1 protein,the distribution of Foxo1 is associated with it's phosphorylation state.Foxo1 protein located in nucleus,after phosphorylated,it was transported from nucleus to cytoplasm obviously;②The expression of FOXO1 gene in mice granule cells is realized through lipofectin transfection;③36h after transfection, the morphology of granule cells begined to change. AnnexinV-PI apoptosis dyeing and Caspase-8 active assay indicated that overexpression of FOXO1 promoted the apoptosis of granule cells and this effect depended on the combination of FOXO1 and DNA;④Also,the results of Real-time PCR indicated that the mRNA level of TRAIL,FasL,Bim,Caspase-3 in granule cells was increased due to FOXOl's overexpression.The experiment confirmed that transcription factor Foxo 1 had an important role in the process of granule cells apoptosis. Foxo1 promoted apoptosis through regulating the expression of TRAIL,Fas L,Bim,Caspase-3 gene directly or indirectly.Thus, Foxo1 may participate in the regulation of follicular atresia.
