| Hu sheep is one of famous multiple birth in the world. The sheep have character of high fertility,estrus at four seasons,early sexal-maturity,multiple birth,heat and humidity tolerance and extensive feeding,fitting confinement feeding and so on.The study that the molecule mechanism of Hu sheep's superfoetation was deeply carry out to develop and utilize hu sheep's resource of polyembryony, It has important significance to breed high-yield variety, improve existent low-yield variety and protect biologic multiformity.Our study mainly showed the expression of Cox-2, Has-2, Tsg-6, Ptx-3 in Hu sheep's various tissue and analyzed their expression levels in the ovary. The results were as follow:1.The study on Cox2 gene expression levels in the ovary of Hu sheepWe designed primer basing on the sequence of Cox2 gene in cattle (GenBank NO.NC-005971). Using the RNA of Hu sheep ovary as template, PCR reaction showed the production was 249bp; The expression profile showed that Cox2 gene expressed in hypothalamus, hypophysis, ovary,oviduct, uterus, heart, liver, spleen, lung, kidney, and muscle. By Real-time PCR the expression of Cox2 gene was investigated. The expression levels of Cox2 in ovaries the group of low Hu sheep were lower than those in high-yield cattle (P>0.05). which proved that the expression level of Cox2 gene mRNA probably had no direct relationship with the reproductive ability of Hu sheep.2. The study on Has2 gene expression levels in the ovary of Hu sheepWe designed primer basing on the sequence of Has2 gene in cattle (GenBank NO.NC-007312). Using the RNA of Hu sheep ovary as template, PCR reaction showed the production was 158bp; The expression profile showed that Has2 gene expressed in hypothalamus, hypophysis, ovary,oviduct, uterus, heart, liver, spleen, lung, kidney, and muscle. By Real-time PCR the expression of Has2 gene was investigated. The expression levels of Has2 in ovaries the group of low Hu sheep were higher than those in high-yield cattle (P<0.01). which proved that the expression level of Has 2 gene mRNA may have a relationship with the reproductive ability of Hu sheep.3. The study on Tsg6 gene expression levels in the ovary of Hu sheepWe designed primer basing on the sequence of Tsg6 gene in cattle (GenBank NO.NC-009161). Using the RNA of Hu sheep ovary as template, PCR reaction showed the production was 197bp; The expression profile showed that Tsg6 gene only expressed in hypothalamus, ovary,uterus, heart. By Real-time PCR the expression of Tsg6 gene was investigated. The expression levels of Has2 in ovaries the group of low Hu sheep were higher than those in high-yield cattle (P<0.01). which proved that the expression level of Tsg6 gene mRNA may have a relationship with the reproductive ability of Hu sheep.4. The study on Ptx3 gene expression levels in the ovary of Hu sheepWe designed primer basing on the sequence of Ptx3 gene in cattle (GenBank NO.NC-007299). Using the RNA of Hu sheep ovary as template, PCR reaction showed the production was 279bp; The expression profile showed that Ptx3 gene expressed in hypothalamus, hypophysis, ovary,oviduct, uterus, heart, liver, kidney, and muscle. By Real-time PCR the expression of Ptx3gene was investigated. The expression levels of Ptx3 in ovaries the group of low Hu sheep were higher than those in high-yield cattle (P<0.01). which proved that the expression level of Ptx3 gene mRNA may have a relationship with the reproductive ability of Hu sheep. |
