Rana "red leg disease" is a major aquaculture industry against Rana, a disease that has a faster onset, spread fast, high mortality characteristics.The traditional method for diagnosis of disease and identification of isolated bacteria, the method is the most reliable method of diagnosis is the standard diagnostic test validation, the typical colonies were found in the culture medium, by smear staining, microscopy, biochemical identification iscan be diagnosed, while at the same sensitivity test done and also reference standard for other diagnostic tests. However, bacterial culture requires a long time, influencing factors, specimen collection and processing of medium components and culture conditions, drawing the location and operation of technology can result in undetected. Therefore need fast, highly specific diagnostic method.In order to rapidly detect pathogens in Yanbian frog red leg disease hydrophila Aeromonas (Aeromonas hydrophila, AH), the test for the detection of frog under the "red leg disease" has established a high specificity and sensitivity of polymerase chaintype reaction (Polymerase Chain Reaction, PCR) and fluorescent antibody technique (indirect fluorescent antibody technique, IFA) collected from the Yanbian region of the frog 32 samples of "red leg disease" to detect infection. Test results show that 32 frog in the Yanbian area, IFA positive number is 14, and the negative number is 18. And PCR, the common frog 14 detected positive and negative 4, PCR positive rate was 87.5% method. IFA law in regard to the sensitivity of PCR,50%(14/28), specificity was 22.22%(4/18), IFA positive detection rate of 43.75% (14/32). The frog "red leg disease" provide the basis for the rapid detection provides.
