Citrus is the second most important fruit in China. Nowadays, the scale of China's citrus industry is larger and larger with an increasing production. So its'market competition becomes more and more fierce. Meanwhile, the quality of our lives has been improved, and we care more about the product quality. Good varieties can take great advantage in the market competition. In this study, Nanfengmiju was used as materials, two samples from liucheng in Guangxi and one from the base ground of Huazhong Agriculture University. AFLP, SRAP, IRAP and other molecular markers were used to identify their differences on DNA level. The genetic relationship of the Nanfengmiju samples from several different places were also discussed here. Additionally, Majiayou, a good pomelo variety, was analyzed. The purity of their seedlings were identified, which did some help for large-scale production. Specific findings are as follows:1,By measuring the leave indexes of the Nanfengmiju samples from Huazhong Agriculture University and liucheng using a vernier caliper, their leaf index are 2.57,1.7 and 2.31.2,120 pares of AFLP primers,156 pares of SRAP primers,56 pares of IRAP primers were used to identify the Nanfengmiju samples from Huazhong Agriculture University and liucheng. The AFLP and IRAP results showed no differences between them. While 6 pares of the SRAP primers could define the differences.3,18 pares of SRAP primers were successfully used to identify the genetic differences between Nanfengmiju samples from 10 different places.4,The different gene sequences were blasted in NCBI database without no matching.5,SSR marker was used to analyze the Majayou and other major pomelo varieties in Guangfeng. The electrophoresis results showed that Majayou had two results, which was dominantly different from the other varieties.
