Ginkgo which is also called white nut is the country level protection plant, and it is the typical mesozoic precious plants, also be called living fossil. Our country has a large number of ginkgo resources in many places. Linchuan and xing'an country are famous hometowns of ginkgo in guiling, Guangxi. There are large areas of ancient ginkgo trees. But now the price of ginkgo biloba is not high and low volume of production, so, many people cut down these ancient trees and sold in other places. So these resources have become much lesser.Microsatellite (SSR) molecular marker technology is the most potential molecular marker; it's also the most advanced genetic markers. This study use six primers to analysis genetic diversity of 13 ginkgo populations from Guilin,and compared with the resources from chengdu. suizhou and han'an, The main results obtain:1. This 6 primers amplification out 34 alleles, each loci for 5.67 alleles, polymorphic loci a percentage to 100%.2. Guilin samples contained only 30 alleles, four alleles have been lost in 34 alleles. They are 166bp gene in GB64 loci and 211 bp,203bp,152bp genes in GB57 loci. And there are four peculiar alleles. It is shows that cultivation of guilin's ginkgo an other regions.3. Alleles are showed be imbalance in 13 populations, except GB65. Each loci has advantages alleles in all samples. Resources from many places have begun to appear the trend of gene purification in some locis.4. Analysis of genetic diversity,13 populations in Guilin are contain alleles different in 20-29, the average number of alleles is 24.92.Shannon index are different in 0.8060-1.3106, the average number is 1.1214.In the genetic diversity degree, expected heterozygosis's numbers are different in 0.4739-0.7096, the average number is 0.6125.By comparison, samples in Guilin show better genetic diversity.5. The analysis shows that, in the genetic variation level,6.4% genetic variation are between population, the genetic variation of individual is 93.6%. The result of molecular variation analysis of variance (AMOVA) also proved this. And the result of diversity comparison with three other population shows there are significant genetic differentiation in different areas.6. From specific allele,genetic diversity level,genetic structure.it's very important to protect of ginkgo resources in Guilin for enrich our ginkgo gene bank.7. According to the results, we propose the protection strategy of Guilin's gingko resources:it is important to protect trees in haiyang town, we can make this place as the base and build a Ginkgo park for protection.
