| Atrazine and chlorpyrifos were widely used herbicides and pesticide, which can be enter the aquatic environment by different ways and eventually accumulate in aquatic organisms, not only can effect plankton, algae and aquatic plants , but also induced toxicological effects on fish. Recent years, toxicity research of them had focused on rodents, on aquatic invertebrates was investigated rarely, in order to abundance the toxicological data, in this study, carp was exposure of atrazine (4.28 , 42.8 and 428μg/L), chlorpyrifos (1.16,11.6 and 116μg/L) and their mixture (1.13,11.3 and 113μg/L), antioxidant level (SOD and MDA ), the related enzyme activities (LDH, ACP, ALP and Na~+/K~+-ATPase), organizational structure, gene expression (HSP70, HSC70, IL-1β, IL-1RI and IFN-γ) and pesticides residues in spleen and head kidney of carp were detced. The results have shown that:1. Atrazine and chlorpyrifos single and joint sub-chronic exposure possible cause of tissue damage, the activity of LDH and ACP was increased and inhibition the activity of ALP and Na~+ /K~+-ATPase in spleen and head kidney of carp.2. The activity of SOD was decreased and the content of MDA was increased in spleen and head kidney by Atrazine and chlorpyrifos and their mixture exposure in spleen and head kidney in common carp.3. Atrazine, chlorpyrifos and their metabolites possible accumulation in the spleen and head kidney tissue of carp after pesticides single and joint sub-chronic exposure.4. Atrazine and chlorpyrifos in the range of concentration, IL-1β, IL-1RI and IFN-γmRNA expression were decreased or increased in spleen and head kidney, the result suggesting that atrazine and chlorpyrifos probably induced immune toxicity by increasing IL-1β, IL-1RI mRNA synthesis in carp.5. The expression of HSP70 and HSC70 mRNA has been dected, we conclude that atrazine and chlorpyrifos induced immunolesion in head kidney and spleen of carp, HSC70 and HSP70 chould proteced the immune cells.In summary, atrazine and chlorpyrifos single and joint sub-chronic exposure, in the appropriate concentration, exposure time and conditions, possible cause cell damage, the activity of SOD, ALP and Na~+/K~+-ATPase was inhibition, the activity of LDH, ACP and MDA was increased, and induce HSP70, HSC70, IL-1β, IL-1RI and IFN-γmRNA expression decreased or increased in spleen and head kidney of carp, which illustrated atrazine and chlorpyrifos had immune toxicity on spleen and head kidney of carp. |
