In this experiment, Inonotus obliquus strains which comes 5 different countries and regions:China, Finland, North Korea, Russia and Japan were used as testing materials to proceed substitute cultivation.Samples were taken 20,40,50,70,85,100, 110,130days after inoculating.And the activity variations of 9 extracellular enzyme including cellulase, hemicellulase, polyphenol oxidase, amylase and protease were determined, on this basis,strain which in China were conducted with GA,KT,2,4-D and 6-BA, the effects of which on mycelial growth, sclerotia formation and extracellular enzyme activity were evaluated. The results is as following:1.The mycelial color,growth vigor,growth rate and sclerotia formation of strains from different countries were various,strains from China,Finland,north Korea produced sclerotia.The dry weight,polysaccharide yield of the strain which in China is much higher than other strains,the biologicalefficiency reached 27%.2.The extracellular enzyme activities in different strains of Inonotus obliquus is:The acivities of CMC and FP enzyme are higher at mycelial growth stage and sclerotia maturity period.The activity variation trends of HC and protease are similar,both of which came to several waves in the whole process of cultivation,however,the activity of protease is much higher than that of HC.The laccase activities of strains differ from each other,with a indication that all of them are high at mycelial growth stage,but higher in 3 strains which could produce sclerotia than the two remaining during the sclerotia formaton period.The activities of polyphenol oxidase,amylase and peroxidase are low,but each of 4 enzymes showed a higher activity in mycelial growth stage than in the sclerotia formaton period apart from the polyphenol oxidase.3 Strains which from China were processed by adding different plant growth regulators and each treatment produced sclerotium.2,4-D promoted the mycelial growth,KT and 2,4-D had significant effects on increaseing the quantity of sclerotium.The polysaccharide yield of 6-BA,2,4-D and KT were notably higher than the the CK, and the activities of protease, CMCase,HCase by 6-BA were higher than others at formation stage and mature stage of sclerotium. In conclusion,the nutrition sources of Inonotus obliquus strains is decomposing substance except lignin in the process of substitute cultivation, while enough lignin nutrition in the formation period of sclerotium is the key factor of sclerotia forming; mycelium growth and amylase activity shows positive correlation; forming sclerotium or not determines by the laccase activity in formation period of sclerotium,the production of sclerotium and activity of protease or cellulase demonstrates positive correlation at the same time shows.
