Polygonatum, the dry root of Polygonatum odoratum (Mill.) Druce, belonging to the Polygonati genus of Liliaceae, is a kind of wild herb with a wide distribution and rich resources in most provinces of China. Its main constituents include polysaccharides, flavonoids, saponins etc. Because of its pharmacological effects and economic benefits, the study of polygonatum odoratum in recent years becomes one of the hot spot in traditional Chinese medicine research.In this research, the HPLC chromatogram of Diosgenin was used as reference peak, chromatography conditions were Welchrom-C8 column. The mobile phase were acetonitrile and water, the analyses were performed by gradient elution, the rate of flow is 1.0ml/min, and detective wavelength is 203nm,the running time is 70min.An HPLC method was established for the fingerprint of polygonatum. The signals of fingerprint were numerically processed. By studying comparatively the fingerprints of 10 samples,9 peaks have been confirmed. The HPLC chromatograms were determined in different places of Polygonatum, different drying methods of polygonatum, different harvest periods and different parts of polygonatum. The relative retention time (RSD) of each peak was less than 3% in the stability, precision and reproducibility study, which revealed that this method was stable and feasible. The similarity is up to 0.9 by Computer Aided Similarity Evaluation software, satisfying the requests of fingerprint. According to hierarchical cluster analysis, the results were basically the same as similarity evaluation.By studying the antioxidant activities of polygonatum odoratum, it was extracted on its different parts and extracted with water. The results showed that polygonatum has some antioxidant activities and the antioxidant activities were different from polygonatum extraction part of water and water extraction. When the concentration got to 100 mg/ml, the scavenging rate of hydroxyl radical about polygonatum extraction part of water and methanol and water extraction reached to 58.88%,53.45%,61.71%, respectively;When the concentration got to 200 mg/ml, the scavenging rate of DPPH·radical and about polygonatum extraction part of water, methanol and water extraction reached to 94.78%,86.28% and 95.32%, respectively.
