The specific primers of the gene tri5 were used to amplify genomic DNA in order to rapidly identify the toxogenic Fusarium strains, which were isolated from the air, feed, soil, dust and water sampled from poultry houses. The condition of toxogenic culture was investigated by orthogonal design method. Quantitative detection of T-2 toxin(HT-2 toxin) was conducted in toxogenic culture product and the samples from poultry houses by immunoaffinity column clean and high performance liquid chromatography(HPLC). The quantitative relationship of T-2 toxin and toxogenic Fusarium strains was analyzed, and their relationship with Fusarium strains from poultry houses was set up into a model to prove the occurrence rule of airborne Fusarium and T-2 toxin in poultry houses. The results provide a theoretical basis for the early hazard warning and control of human and animal fungal diseases.Totally 19 airborne samples and 164 substrate samples including soil, grain, feed and water were collected from poultry houses in November 2009 and April 2010 in a poultry houses in Dalian. After fungal isolation and culture, 59 Fusarium isolates from airborne and 80 Fusarium isolates from substrate were identified. Among which, F. verticillioides (23.7%) and F. semitectum (17.26%) were dominant strains. By comparison with other substrates, the concentration of Fusarium in soil was highest. Most Fusarium strains were isolated from chicken house. 42 tri5 positive strains were screened by tri5-PCR and the dominant tri5 positive Fusarium species was F. graminearum. The highest percentage of tri5 positive strains was isolated from soil. Toxogenic culture of 10 tri5 positive Fusarium was from airborne according to the optimal toxogenic culture condition, and the product was detected by HPLC. Seven Fusarium strains produced T-2 toxin, their concentrations were 1.3ng/mL~5ng/mL, while three Fusarium strains produced HT-2 toxin with the highest concentration of 17.1ng/mL.According to the quality testing standard of the National Quality Inspection Administration of China, aflatoxin(AF), zearalenone(ZEA), ochratoxin A(OTA) and T-2 toxins were detected by immunoafinity column clean-HPLC. T-2 toxin was obtained in all kinds of substrates and the concentration in soil was the highest (21μg/kg). OTA and ZEA were not checked out in samples except feed. The concentration of ZEA in spring was significantly different from that in winter. AF was undetcted.
