| Chestnut flowers of Varieties of yanshan were selected as raw materials fortesting in this paper. The extraction and purification of flavonoids in chestnut flowerswere systematically studied. The main results are as follows:(1) The determination of total flavonoids in chestnut flowersThe spectrophotometry to determination of the content of total flavonoids inchestnut was built with rutin as standard control sample. It was proved that themethod was rapid and accurate through reproducible test, stability test and therecovery test. It provided a scientific evaluation method to explore the indicators ofoptimum extraction and purification process.(2) Extraction process of total flavonoids in chestnut flowersAccording to the extraction rate of total flavonoids,on the basis of single factorexperiment, it obtained the optimum conditions for the two methods by orthogonaldesign and variance analysis. The optimal conditions of Microwave-assistedextraction: ethanol concentration was50%, material liquid ratio was1:90, themicrowave power was800W, and extraction time was82s. Under these conditions,the extraction rate of total flavonoids of chestnut flowers was about3.49%. Theoptimal conditions of CO2supercritical extraction:material liquid ratio was1:7.5,extraction temperature was45℃, extraction pressure was40MPa, and extractiontime was1.0h. Under these conditions, the extraction rate of total flavonoids ofchestnut flowers was about1.116%. The test extracted and determined the totalflavonoids of chestnut flowers of different flowering ands different organs of chestnutflowers by microwave-assisted extraction method.(3) Purification and analysis of total flavonoids in chestnut flowersBy screening for the five kinds of macroporous resin, D101, X-5, NKA-9, AB-8and XAD-2, it was proved that AB-8type resin was suitable for purification offlavonoids in chestnut flowers. Through the research on the purification of flavonoidsby AB-8type resin, it initially obtained the optimum conditions for the AB-8resin purification of total flavonoids in chestnut flowers. The purified extract of chestnutflowers was analyzed by mass spectrometry technique.(4) Research on volatile compounds of Chinese chestnut flowersThe volatile compounds were extracted by CO2supercritical extractiontechnology. By single factor experiment and orthogonal experiment, it obtained theoptimum technological conditions: extraction temperature was40℃, extractionpressure was15MPa, and extraction time was2.0h. Under these conditions, theextraction rate of volatile compounds in chestnut flower was about1.116%. Theextracts were analyzed and identified by gas chromatography-mass spectrometrytechnique.40compounds were identified and29kinds of compounds were detectedfor the first time in the chestnut flowers. |
