In this paper, the rhizoma of Menispermum dauricum DC. commonly called "Bei Dougen" was taken as the object of the study, using MTT assay to screen the extraction process of Bei Dougen. The fingerprint of ten batchs of Bei Dougen from different sources was constructed by HPLC and the inhibition of cell proliferation was evaluated by the cytotoxic activity experiment (MTT). The relationship between the "fingerprints" and "efficacy" was constructed by using correlation analysis and multiple linear regression analysis to explore the material foundation of pharmacological activity.In the experiment, Bei Dougen was processed by eleven methods including warm immersion, ultrasonic and reflux methods according to the principle of acid extraction alkali precipitation. Then, we get the Bei Dougen extracts by different extraction process. The human breast cancer cell line MCF-7was chosen to screen Bei Dougen extract with the MTT. Therefore, we can determine the best extraction methods. The fingerprint of ten batches of different sources of Bei Dougen extract was established by using high performance liquid chromatography (HPLC). In the fingerprint, we can evaluate the similarity of fingerprint by choosing eight common pesk and classify the ten batches of Bei Dougen extracts from different sources by using cluster analysis. Finally, on the basis of the data of the HPLC fingerprint and its cytotoxic activity, the fingerprint-efficacy relationship between HPLC fingerprint and cytotoxic activities were investigated using multiple linear regression (MLR) and correlation analysis.
