Because collagen has some advantages of hemostasis, promoting cell growth, biodegradability, so it can be used to prepare hemostatic dressings. To guard against wound infection, we can add antibacterial agent Ag4O4 to the dressing. Now the common collagen products are mostly extracted from terrestrial species. However, recently people began to doubt the safety of collagen of terrestrial animals sources. Moreover, terrestrial animals are limited in islamic region. Therefore, it is significant to exploit fish collagen. As a freshwater fish, Mylopharyngodon piceus(also called Blakcarp or Herring) is easy to be feed, and it is a kind of important economic fish in China. In addition, the herring is very large, and rich in collagen. For this purpose, this paper studies on the following:1. Extraction technology of collagen. First, the research explored the collagen from the skin, fish scales, swim bladders of herring, by the acid method and enzymatic method, and obtained acid soluble collagen (ASC) and enzyme soluble collagen (PSC), correspondingly. Next, we determine extraction program that extracted PSC from the swim bladders of herring by enzymatic. In addition, this paper discussed and optimized the experimental scheme, the extraction rate of collagen reached up to 8.67%; Finally, in order to use the collagen effectively, the collagen was characterized by some testings, such as hydrogen NMR spectrum, infrared spectrum, SDS-PAGE electrophoresis, differential scanning calorimetry, and so on. We can see that the purity of collagen is very high, the collagen has whole three spiral structure, the molecular weight of collagen is about 100kDa-200kDa, the denaturation temperature of collagen is 40~60℃. Therefore, the collagen from Mylopharyngodon piceus has the potential to take place of that from terrestrial animals.2. Electrostatic spinning process of collagen. After a lot of experiments, the paper found the optimum condition of the electrospinning, which is 5% collagen(mass fraction, solvent is Hexafluoroisopropanol),20 kV voltage,12cm,0.6 ml/h. In that condition, we can obtain even thickness collagen fibers with about 100 nm average diameter. After crosslinked by 25% glutaraldehyde steam, the chemical property of collagen has no change except for hydrophobic increasing. So electrostatic spinning is a good method to prepare biological material.3. The preparation and characterization of Ag4O4. At first, we study out the best experiment scheme by soft chemistry technique. Then product properties were discussed by x-ray diffraction, raman scattering, thermogravimetric analysis, and scanning electronic microscope etc. Finally, the results of quantitative and qualitative antimicrobial tests showed that 0.1% Ag4O4 (ammonia water) has significant antimicrobial effect against e. coli, staphylococcus aureus, candida albicans and pseudomonas aeruginosa.4. Added Ag4O4 to the collagen fibers. The collagen fibers dressing combined with the antibacterial agent not only stopped bleeding quickly, but also had good antibacterial effect against the representative pathogenic bacteria, such as e. coli, staphylococcus aureus, candida albicans and pseudomonas aeruginosa. From above, the material we prepared can be used as a biomedical dressings with antibacterial hemostatic function.
