Tobacco is one of the special economic crops and the main purpose is smoking. aroma taste is the important factor of tobacco quality, the high-quality tobacco requires pure aroma, moderate strength, mellow taste, clean and comfortable surplus flavor and tasting safely. The chemical constitutes of tobacco have a direct influence on the quality of tobacco products. Tobacco alkaloids are the most important chemical composition in tobacco. which is the quality element and characteristic of tobacco products., in the process of cigarette, researches are focusing on the variation regulation of alkaloids in tobacco with the study of Chinese special art deeply. A method of the rapid determination of six alkaloids in tobacco and cigarette mainstream smoke was developed using ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). And analysising the variation regulation of alkaloids in the process of redrying art and primary art. The summary as follows:1. For the purpose of understanding the content level and composition of alkaloids. A method of UPLC-MS/MS alkaloids was established which was convient and quick to analysis the main alkaloids in tobacco.And this method was also used to analysis the main alkaloids in tobacco products. The results indicated that:â‘ Nicotine in flue-cured tobacco is much higher than the other alkaloids;â‘¡In flue-cured tobacco, anatabine, nornicotine and anabasine was much lower, the content of myosmine and cotinine were the lowest alkaloids in the six determined alkaloids, which belongs to the micro alkaloids;â‘¢The content of alkaloids in tobacco followed the sequence:nicotine> anatabine> nornicotine> anabasine> myosmine> cotinine.2. In order to study the general knowledge about the content and transfer rule of alkaloids in mainstream smoke, a method of UPLC-MS/MS was established which was convient and quick to analysis the content of six main alkaloids in tobacco. Results show that:â‘ The mobility of nicotine, nornicotine and anabasine were 3.83%-12.16%,0.89%-7.69% and 0.91%-6.85% respectively;â‘¡The mibility of anatabine is in the range of 0.30%-1.10%;â‘¢The mobility of myosmine and cotinine were much higher, which was 11.08%-95.98% and 2.31%-31.87%. This indicated that parts of myosmine and cotinine in mainstream smoke was formed from the transformation of alkaloids in burning cigarette.3. This established new method was used to analysis the variation regulation in three key production processes, that was first casing, second casing, and drying in redrying process and three or more key production processes, that was loosening and humidifying, weting and casing, and drying in primary process. The result indicated:â‘ In redrying process, the influence of production processes to alkaloids was in the follow sequence:the drying process> first casing process> second casing process;â‘¡In primary process, which followed the sequence:high-temperature drying process> the loosening and humidifying process> the weting and casing process.
