To sear intraspecific taxonomy and the levels of genetic diversity of Rhinolophus ferrumequinum from some areas around Henan Province, The dissertation studies Rhinolophus ferrumequinum from eleven sampling sites of four privinces (Hubei, Anhui,shanxi and Hebei Province) in morphological difference, we analyse the different eleven populations of Rhinolophus ferrumequinum by morphological and molecular biology. First, we measure the morphological charters of Rhinolophus ferrumequinum, and analies by SPSS (Statistical Package for the Social Science or Statistic Products and Service Solution)13.0, to verify the difference on the morphological charters and identify its intraspecific taxonomomic status. In addition, we amply mitochondrial cytb gene, digest amplified products of mtDNA cytb gene by restriction endonucleases, use molecular biology softwarest to analysis mitochondrial(mt) cytb gene sequence and restriction endonucleases results, to rearch intraspecific taxonomomic status, which show the levels of genetic diversity of Rhinolophus ferrumequinum and phylogenetic relationtionships among geographic populations. The results as:Neighty-seven Rhinolophus ferrumequinum were studied from eleven sampling sites of four privinces (Hubei, Anhui,shanxi and Hebei Province) in morphological difference.we apply morphological data and analyse by cluster anapysis, PCA,discriminant anapysis and One Way ANOVA. The results showed: morphological charters of 11 geographic populations have some difference,but the difference is not visible.Studying phylogenesis of eleven geographic populations of Rhinolophus ferrumequinum by molecular biology,we amply the mt Cytb gene, construct phylogenetic tree, the results showed: 11 geographic populations of Rhinolophus ferrumequinum and Japan population are clustered in the same clade,Yunnan samples cuuster alonely. The geographic populations ranged from 0.0% to 2.1%. We think Rhinolophus ferrumequinum of study area is R. f. nippen Temminck, and different geographic populations appeared to genetic variation.RFLP(Restriction fragment length polymorphism) technique are used to analyse genetic diversity. Five restriction endonucleases were used to analyze the amplified products of mtDNA cytb gene. The results showed: The Cytb gene was digested by BamH, HaeⅢand Hinf I which produced the same restriction sites and this three restriction endonucleases don't have no RFLP appears. In contrast, RFLP were found products in the digested by TaqⅠand AfaⅠfor three geographic populations.So,this three geographic populations maybe have genetic diversity.
