The interaction research of protein and metal ions is an important part of the life science and technology field; it is the focus research in the chemical and the life science fields in the past ten years. Serum albumin is the most important spherical protein in the animal plasma, and is the carrier protein of many metal ions. Bovine serum albumin is quite similar to the human serum albumin in the structure, and is widely studied as a model protein. Lysozyme presents in various tissues of living body, and is contained most abundant in egg white. Hen egg white lysozyme consists of 18 species and 129 amino acids, and is a single peptide chain. Because of its low molecular weight, lysozyme is one of the most important model proteins in the protein structure research. Copper ion, zinc ion and calcium ion are the metal ions that cannot be lacked in the life system and normal function playing of the living body.In recent decades, people study the interaction of the metal ions and the protein via all kinds of research tools and analysis methods. Two-dimensional correlation infrared spectroscopy technology is a powerful and flexible spectroscopy analysis technology. In the research of protein structure and conformation relating fields, it shows unique advantages in the intractable problem that the protein has a correspondingly weak infrared absorption and some wide peaks or lapped peaks are difficult to distinguish.In this thesis, preliminary analysis and research on the interaction of the metal ions and the protein are done via the method of Fourier transform infrared spectroscopy.(1) Studying the various instrument setting conditions and experimental conditions in the infrared spectroscopy measuring and data processing methods. Choosing heavy water as the solvent, time as the outer perturbation, H-D exchanging as the studying mode and two-dimensional correlation infrared spectroscopy technology as the main analysis means. Collecting the relating information on the interaction of the metal ions and the protein via infrared data and doing the preliminary analysis.(2) Analyzing the infrared data on the interaction of the metal ions and bovine serum albumin. The analysis result reveals that three metal ions, copper ion, zinc ion and calcium ion, obviously affect the bovine serum albumin deuterated process and the secondary structure deuterated sequence(3) Analyzing the infrared data on the interaction of the metal ions and lysozyme. The analysis result reveals that three metal ions, copper ion, zinc ion and calcium ion, obviously affect the deuterated degree of lysozyme, while have no influence on lysozyme deuterated process and the secondary structure deuterated sequence.
